Identification and characterization of a serotype b-specific segment of the Haemophilus influenzae genome

Abstract: Within the region of the Haemophilus influenzae serotype b genome required for capsule expression, a 4.6-kilobase segment was cloned, used as a hybridization probe, and found to be specific for serotype b strains. Deletion mutagenesis within this segment defined a locus necessary for serotype b capsule production and another involved in maintenance of wild-type colony appearance.

“…Both bwas performed in which control and infected tissue were variants (RM2177 and RM132) caused accumulation of deincubated and examined by light microscopy, TEM, and bris and extrusion of epithelial cells. Examination of serial SEM after 30 min, 4 h, 14 h, and 24 h (except for explants 1-,um sections revealed damage that varied from localized infected with Ela/R1369, for which SEM was not conduct-(patchy) to generalized (widespread) loss of epithelial integed).…”

“…Strain RM2177 has noniridescent colonies, does not react with antiserum to H. influenzae type b, lacks polyribosyl-ribitol phosphate in culture supernatants, and lacks surface immunofluorescence when reacted with fluorescein-conjugated rabbit antiserum to polyribosylribitol phosphate. It is also avirulent in the infant rat model of systemic H. influenzae infection (4). RM132 is a derivative of strain Rd, a btype d strain (32).…”
