“…The high proportion of S2 isolates with a nonsynonymous mutation at the corresponding codon indicates great genetic instability of this serotype, in accordance with data from several previous reports (5,22,23). Most S3 isolates were mutated at T 6 (T3I), but the value of this attenuation marker is controversial (3,32,33), as such substitutions have been found in three substrains of OPV that were used without any reported adverse reactions (28).…”

“…Among the mutations observed in the VP1 regions, special attention was paid to nonsynonymous mutations affecting codons of amino acid residues that are known to be implicated in the attenuation phenotype of OPV strains (attenuation markers): VP1 residues A 88 , T 106 , and F 134 of the serotype 1 Sabin strain (S1) (2); VP1 residue I 143 of S2 (22); and VP1 residue T 6 of S3 (32,33) (Fig. 2).…”

“…143 in S2 (32), and T 6 in S3 (33,34). In the VP1 sequence of the 23 S1 isolates, we found 12 isolates with an amino acid substitution at T 106 (52%) (Fig.…”

“…Thus the largest part of the attenuated phenotype of the type 3 Sabin vaccine strain of poliovirus was attributable to only two mutations. Subsequently a third mutation at nucleotide 2493, which is believed to have an attenuating effect, was identified by other workers, leading to an amino acid change from isoleucine to threonine at residue 6 of capsid protein VP 1 (Weeks- Levy et al, 1991 ;Tatem et al, 1992). This difference was not observed in our original clones probably because the virus from which they were derived has been plaque-purified and passaged twice in HEp2C cells, and it has been shown that the mutation is rapidly lost on passage.…”
