Identification and biochemical characterization of a novel endo-type β-agarase AgaW from Cohnella sp. strain LGH

Abstract: An agar-degrading bacterium, strain LGH, was isolated and identified as Cohnella sp. This strain had a capability of utilizing agar as a sole carbon source for growth and showed a strong agarolytic activity. A novel endo-type β-agarase gene agaW, encoding a primary translation product of 891 amino acids, including a 26 amino acid signal peptide, was cloned and identified from a genomic library of strain LGH. The AgaW belonged to the glycoside hydrolase (GH) GH50 family, with less than 39% amino acid sequence s… Show more

“…Over and above, some of the metal ions had inhibitory or negative effects on the enzyme activity of the agarases. The partial inhibition of agarase activity by Ba 2+ ions was detected in AgaW (Li et al 2015). The Sr + and Ba 2+ ions inhibited the activity of Aga4436 (Chen et al 2016).…”

“…A handful of metal ions positively influenced the activity of a few β-agarases. The redox reagent dithiothreitol (DTT) had stimulatory effects on the agarases AgaB, AgaW, AgaXa, AgaM1, and HZ2 (Dong et al 2016;Li et al 2015;Xie et al 2013;Di et al 2018;Lin et al 2012). At higher concentrations of 9 mM and 10 mM, DTT increased the activity of HZ2 and AgaXa respectively (Lin et al 2012;Xie et al 2013).…”
