Icariin prevents cytokine‑induced β‑cell death by inhibiting NF‑κB signaling

Abstract: Abstract. The loss of insulin secretion in type I diabetes mellitus (T1DM) is caused by autoimmune-mediated destruction of insulin-producing pancreatic β-cells. Inflammatory cytokines and immune cell infiltration activate oxidative and endoplasmic reticulum (ER) stress, resulting in reduced β-cell viability. The current pharmacological agents used to control blood glucose have a limited effective duration and are accompanied by strong side effects. Blocking the inflammatory and immune responses that cause the … Show more

“…ICRN (C 33 H 40 O 15 , M w = 676.67) is a type of prenylated flavonoid, showing extensive bioactivities such as antioxidant [ 44 , 45 , 46 , 47 ], neuroprotective [ 48 , 49 , 50 , 51 , 52 ], and antitumor [ 53 , 54 , 55 , 56 , 57 , 58 , 59 , 60 , 61 ] behavior as well as anti-inflammatory responses [ 62 , 63 , 64 , 65 , 66 , 67 , 68 ], and can be used to treat erectile dysfunction [ 69 , 70 , 71 , 72 ]. It appears it can improve the function of organs including bones and the heart [ 73 , 74 , 75 , 76 , 77 , 78 , 79 , 80 , 81 , 82 , 83 , 84 , 85 , 86 ].…”

“…In vitro, baicalin decreased the apoptosis of the mice pancreatic β cells that were treated with TNF‐α by upregulating miR‐205 resulting in activating the PI3K/Akt pathway and inhibiting the NF‐κB pathway 129 . Icariin, a flavonoid glycoside, prevented the death of cytokine‐induced rat pancreatic β‐cell (RINm5F cells) by inhibiting NF‐κB signalling 130 . Moreover, genistein significantly decreased the incidence of T1DM in female non‐obese diabetic mice 131 .…”

“…The quantification of the nuclear translocation of NF-κB has been established by using high-content fluorescence microscopy and imaging flow cytometry [57]. On the other hand, EMSA [37] and commercial DNA binding ELISA (such as TransAM ® NF-κB activation assays from Active Motif ® ) have been widely used to assay NF-κB activation [58][59][60][61].…”
