Ibotenic acid-induced lesions of the medial zona incerta decrease lordosis behavior in the female rat.

Abstract: To examine the role of the medial zona incerta (m/I) in female sexual behavior, ovariectomized estrogen-and progesterone-treated female rats were tested for sexual receptivity after bilateral injections of the selective neurotoxin ibotenic acid (3 jig/0.3 id) directly into the mZI. These injections produced a significant attenuation of lordosis behavior in highly receptive females when compared with saline-injected controls. This decrease in sexual receptivity was also reflected in a significant increase of re… Show more

“…The IHDA neuronal system is a diverse one that comprises DA neurons in the medial zona incerta (A13) and rostral hypothalamic periventricular nucleus and their projections within and outside the hypothalamus (4,11,27). The functional role(s) played by the IHDA system has not been ascertained, although it may involve the neuroendocrine control of luteinizing hormone secretion (18,19,38), as well as sexual (10) and ingestive (43) behaviors in rats. Earlier studies (16,42) indicate that IHDA neurons behave like the NSDA and MLDA neurons in their responses to opioids, i.e., being Half of each group received injection of either OFQ (1 g icv) or vehicle in the morning on day 6 and were decapitated 60 min later.…”

“…Although progestin receptors were found in many dopaminergic cell groups after oestradiol treatment, we found no evidence of progestin receptor expression in dopaminergic cells of the zona incerta, consistent with previous reports in female rats (10, 11), guinea‐pigs (17) and macaques (18). The dopamine projections from the zona incerta have been studied for a role in the control of gonadotropin release necessary for ovulation (19), as well as for sexual receptivity in female rats (20). However, it is not clear if progesterone is capable of altering dopaminergic activity of the zona incerta (19–23).…”

However, it is not clear if progesterone is capable of altering dopaminergic activity of the zona incerta (19–23). Even if progesterone does affect dopaminergic activity in the zona incerta, the absence of progestin receptors in dopaminergic cells suggests that it does not do this by binding to intracellular progestin receptor within these cells.…”

“…The IHDA neuronal system comprises those dopamine neurones in the medial zona incerta (A13) and rostral periventricular hypothalamic regions (A14), and their local projections within the hypothalamus ( 41, 42). The IHDA system, specifically that in the zona incerta, may be involved in the control of LH secretion and ovulation ( 43–45), lordosis behaviour ( 46), water and food intake ( 47) in rats. We showed in this study that cysteamine significantly increased both DOPAC and dopamine concentrations in the paraventricular nucleus and A14 (Fig.…”
