Ia determinants on human T-cell subsets defined by monoclonal antibody. Activation stimuli required for expression.

Abstract: The nature of Ia antigens which appear on human T cells after activation and the stimuli required for their expression was examined utilizing a monoclonal antibody reactive with the Ia antigen framework. T cells were purified using monoclonal antibodies directed either at the entire T-cell population (OKT3) or the T-cell inducer subset (OKT4). By indirect immunofluorescence, it was shown that the human T-cell population contains no detectable Ia+ cells in the resting state. In contrast, in excess of 60% of the… Show more

“…Nonetheless, HLA class II molecules were up-regulated in activated B cells from YAC-transgenic mice (data not shown). Second, in humans, but not in mice, class II molecules are expressed on activated T cells (41). Accordingly, expression of DR and DQ was not detectable on mouse T cells (Fig.…”

“…It has been demonstrated that some cell surface molecules such as CD69, CD25, ICAM-1 and HLA-DR are up-regulated following T cell activation and differentiation (43)(44)(45)(46). Given that TLR2 engagement leads to homotypic aggregation of quiescent naive and memory CD4 ϩ T lymphocytes, we next assessed whether treatment with the TLR2 agonist Pam 3 Csk 4 can increase expression of some specific cell surface markers.…”

“…This antibody lacked specificity for unstimulated lymphocytes. la antigens, expressed primarily on B lymphocytes or monocytes, are found in a small percentage of freshly isolated T cells, but in significant proportions of activated T cells (26)(27)(28). Recently a monoclonal antibody termed anti-Tac has been described that is reactive with a proportion of human T cells activated by mitogens, soluble antigens, or alloantigens, but not resting T cells, B cells, or monocytes (29).…”
