ʲ-mannanase of Streptomyces lividans 66: cloning and DNA sequence of the manA gene and characterization of the enzyme

Abstract: The gene coding for a beta-mannanase was cloned homologously from Streptomyces lividans and its DNA sequence was determined. The fully secreted enzyme was isolated and purified from culture filtrates of the hyperproducing clone S. lividans IAF36 grown in mineral salt media containing galactomannan as the main carbon source. It had a molecular mass of 36 kDa and a specific activity of 876 i.u./mg of protein. Under the assay conditions used, the optimal enzyme activity was obtained at 58 degrees C and a pH of 6.… Show more

“…b-1,4-Mannanases (E.C.3.2.1.78), catalyzing the hydrolysis of the b-1,4 glycosidic linkages in mannan, galactomannan, glucomannan and galactoglucomannan [4,5], have been purified and characterized from several different sources such as plants, fungi and bacteria [6][7][8][9][10][11]. For some of the b-mannanases, the cDNA sequences have been determined.…”

“…The three conserved active site residues (Asn-121, Glu-122, and Glu-223) and the strictly conserved residues Arg-43, His-80, His-188, Tyr-190, and Trp-248 that are characteristic of all family 5 enzymes (2, 4, 20, 28, 49) were also found in ManAd3. The mannanase domain of ManA exhibited 71% sequence identity with the mannanase catalytic domains of ManA and CelC of Caldicellulosiruptor saccharolyticus (19,33) and 56% sequence identity with the ␤-mannanase ManA of Streptomyces lividans (1).…”

“…In contrast, two or more genes classified as members of the GH74 family in the CAZy database are present in many bacteria. Streptomyces species are the most extensively studied of these bacteria and have been used as a source of hemicellulases (2,4,9,12,19,20,23,28,32). Streptomyces avermitilis possesses two GH74 genes, which are similar to those found in Geotrichum sp.…”
