Watermelon chlorotic stunt virus from the Sudan and Iran: Sequence Comparisons and Identification of a Whitefly-Transmission Determinant

Abstract: The genomes of two Watermelon chlorotic stunt virus (WmCSV) isolates, one from the Sudan and one from Iran, were cloned and sequenced. Sequence relationship with other geminiviruses characterizes WmCSV as a typical Eastern Hemisphere geminivirus with a bipartite genome. The two geographically distant WmCSV isolates from Africa and the Middle East share a very high overall sequence similarity: 98% between their DNA-A and 96% between their DNA-B components, and their respective capsid proteins are identical. A s… Show more

“…3b) showed that the BotGn-1 belonged to the ToLCNDV cluster. Symptoms of the disease are similar to watermelon leaf curl disease described from Pakistan [18] and watermelon chlorotic curly stunt disease described from Iran and Sudan [5,13] which also result in mottling, curling of leaves and stunting of plants. The virus associated with the CCS disease of bottle gourd did not infect watermelon by whitefly inoculation, indicating differences between the viruses or their strains causing these diseases.…”

“…Since then, the diseases caused by begomoviruses have been reported in bitter gourd, chayote, cucumber, muskmelon, pumpkin and sponge gourd [17,20,22,30,34]. Many different begomoviruses infecting cucurbits have been reported from other countries such as Squash leaf curl Yunnan virus from China [39], Melon chlorotic leaf curl virus from Guatemala [3] Chayote yellow mosaic virus from Nigeria [33], ToL-CNDV from Thailand [26,27], Watermelon chlorotic curly stunt virus from Sudan and Iran [13], Squash leaf curl Philippines virus from the Philippines [14], Cucurbit leaf curl virus [2], Cucurbit leaf crumple virus [9], Squash leaf curl virus [8,16] and Squash mild leaf curl virus [4] from USA, and Luffa yellow mosaic virus from Vietnam [24].…”

“…The single CP possesses a molecular mass of 30.1 kDa and a calculated pI of 10.0. Each geminate particle encapsidates one circular DNA molecule so that two virions are needed for infection in case of bipartite geminiviruses.Encapsidating the viral genome, the geminivirus CP is important for insect transmission (Briddon et al, 1990; Höfer et al, 1997; Höhnle et al, 2001;Kheyr-Pour et al, 2000; Noris et al, 1998) and transport of the viral DNA in and out of the nucleus (Kotlizky et al, 2000;Kunik et al, 1998Kunik et al, , 1999 Liu et al, 1999;Unseld et al, 2001Unseld et al, , 2004. For the ACMV CP, three regions have been determined that mediate nuclear import (Unseld et al, 2001), whereby the central region is not only important for nuclear import but also for export.…”

“…All cucurbit-infecting begomoviruses reported at present have a bipartite genome (18). In the Eastern Hemisphere, Watermelon chlorotic stunt virus has a native range spanning Sudan, southern Arabia, and Iran (23,33). (25) occurs in China, the Philippines (34), and Vietnam (34), whereas Squash leaf curl Yunnan virus has been reported only in southern China (54).…”
