Sox17expression in endocardium precursor cells regulates heart development in mice

Abstract: The endocardium is the endothelial component of the vertebrate heart and plays a key role in heart development. Cardiac progenitor cells (CPCs) that express the homeobox gene Nkx2-5 give rise to the endocardium. Where, when, and how the endocardium segregates during embryogenesis have remained largely unknown, however. We now show that Nkx2-5 + CPCs that express the Sry-type HMG box gene Sox17 specifically differentiate into the endocardium in mouse embryos. Approximately 20% to 30% of Nkx2-5 + CPCs transientl… Show more

“…Lineage tracing of early Notch1+ cells preferentially marks the endocardium, suggesting a potential role for the Notch pathway in early CPC fate decisions between CMs and ECs (Lescroart et al 2018). Moreover, further crosstalk between these two lineages happens in vitro and in vivo, suggesting paracrine mechanisms governing CPC specification and differentiation (Saba et al 2019;Saint-Jean et al 2019). Interestingly, endocardial cells with "valve-like" properties can be derived from hPSCs through the activation of other signaling pathways.…”
