HLA‐DRB1*15:01:43 and HLA‐DRB1*15:01:44 alleles were identified by next‐generation sequencing

Abstract: submitted to the IPD-IMGT/HLA Database 2 in December 2021. This follows the agreed policy that, subject to conditions stated in the most recent Nomenclature Report, 3 names will be assigned to new sequences as they are identified. Lists of such names will be published in the following WHO Nomenclature Report. ACKNOWLEDGMENTSThe authors would like to offer special gratitude to the volunteer donors for their participation in our bone marrow donor registry. We would also like to thank Dharma Master Cheng Yen, fou… Show more

“…HLA‐DRB1*08:03:13 allele was found in the donor from the Cord Blood Bank of Zhejiang Province, China. The sample was genotyped for HLA‐A, ‐B, ‐C, ‐DRB1, ‐DQB1, and ‐DPB1 loci by NGS with AllType™ 11‐loci kit (One Lambda Inc., Canoga Park, CA) as previously reported 2,3 . HLA‐DRB1*12:02 allele and a novel allele, now named as HLA‐DRB1*08:03:13 were identified in the sample.…”

The sequence of exons 2 to 6 of the HLA-DRB1*08:03:13 allele was identical to that of the HLA-DRB1*08:03:02:01 allele, except for one nucleotide change at position 309 G>A in exon 2, which is a synonymous change that does not lead to amino acid change (Figure 1 The names HLA-DRB1*08:03:13 and HLA-DRB1*08 :119 have been officially assigned by the WHO Nomenclature Committee for Factors of the HLA System in January 2022 and December 2022 respectively.…”

“…The proband comes from the Cord Blood Bank of Zhejiang Province, China. The sample of the proband was firstly genotyped for the HLA‐A, ‐B, ‐C, ‐DRB1, and ‐DQB1 loci at high resolution level using the AllType™ NGS 11‐loci amplification kit (One Lambda Inc., Canoga Park, CA, USA) and the genotype of the sample was assigned with TypeStream Visual Software version 2.0(One Lambda Inc.) as previously reported 2–4 …”

[2][3][4] The novel allele HLA-C*01:220 was identical to HLA-C*01:02:01:01 except for one single nucleotide substitution A to T at position 559 in exon 3, which resulted in an amino acid change from Threonine to Serine at residue 163 (Figure 1). The accession number OK106260 in the GenBank nucleotide sequence database was assigned for the nucleotide sequence of the HLA-C*01:220 allele.…”
