28Type 2 diabetes mellitus (T2DM) is a chronic metabolic disease that is becoming a 29 significant global health care problem. Several studies have shown that people with 30 diabetes are more susceptible to oral problems, such as periodontitis and, although the 31 causes are still inconclusive, the oral microbiota seems to be an important factor in this 32 interaction. This study aimed to characterize the oral microbiome of a sample representing 33 T2DM patients from Portugal and exploit potential associations between some 34 microorganisms and variables like teeth brushing, smoking habits, and nutrient intake. 35 By sequencing the hypervariable regions V3-V4 of the 16S rRNA gene in 50 individuals 36 belonging to a group of diabetes patients and a control group, we found a total of 233 37 taxa, from which only 65% were shared between both groups. No differences were found 38 in terms of alpha and beta diversity between groups or habits categories. Also, there were 39 no significant differences in the oral microbiome profiles of control and diabetes patients. 40Only the class Synergistia and the genus TG5, which are related to periodontitis, were 41 statistically more frequent in the control group. This finding can be justified by the fact 42 that these diabetic patients usually have their oral health under close medical control than 43 an average healthy person, which in this study was represented by the control group. 44 45 IMPORTANCE: Diabetes has become a significant global health care issue as its 46 incidence continues to increase exponentially, with type 2 diabetes being responsible for 47 more than 90% of these cases. Portugal is one of the countries with a higher prevalence 48 of diabetes in Europe. It has been reported that diabetic people have an increased risk of 49 developing several health problems such as oral infections mostly caused by opportunistic 50 pathogens. Some studies have pointed out a relationship between diabetes and oral 51 3 microbiome. Therefore, the characterization of the microbial ecosystem of the mouth in 52 reference groups is crucial to provide information to tackle oral health pathogen-borne 53 conditions. Tongue diagnosis is a unique method in traditional Chinese medicine (TCM). This is the first investigation on the association between traditional tongue diagnosis and the tongue coating microbiome using next-generation sequencing. The study included 19 gastritis patients with a typical white-greasy or yellow-dense tongue coating corresponding to TCM Cold or Hot Syndrome respectively, as well as eight healthy volunteers. An Illumina paired-end, double-barcode 16S rRNA sequencing protocol was designed to profile the tongue-coating microbiome, from which approximately 3.7 million V6 tags for each sample were obtained. We identified 123 and 258 species-level OTUs that were enriched in patients with Cold/Hot Syndromes, respectively, representing "Cold Microbiota" and "Hot Microbiota". We further constructed the tongue microbiota-imbalanced networks associated with Cold/Hot Syndromes. The results reveal an important connection between the tongue-coating microbiome and traditional tongue diagnosis, and illustrate the potential of the tongue-coating microbiome as a novel holistic biomarker for characterizing patient subtypes.

Aim: To perform a comparative analysis between two methods for detecting Porphyromonas gingivalis, Tannerella forsythia and Porphyromonas endodontalis in periodontal plaque samples. Methods: The study sample consisted of twenty systemically healthy patients showing generalized chronic periodontitis. The subgingival samples for microbiological analysis were collected before (baseline) and 60 days after a basic periodontal therapy from 30 non-adjacent affected sites (Probing Depth (PD): 5-7 mm, Clinical Attachment Loss (CAL) ≥ 5 mm, positive for Bleeding on Probing (BOP)). Microbiological analysis was performed by PCR and qPCR. To allow a comparative analysis between both methods, qPCR was divided in three different scores (score 2: presence of more than 100 bacteria; score 1: presence of 10-100 bacteria, and score 0: absence of bacteria), in accordance to DNA quantity, while for PCR two scores were assigned: presence or absence of bacteria. Results: qPCR demonstrated higher sensitivity in the detection of these pathogens compared with PCR when scores 1 and 2 were considered positive. However, when only score 2 was considered positive, PCR and qPCR showed better agreement. Conclusions: qPCR demonstrated higher sensitivity than conventional PCR for detection of low numbers of microorganisms and can be useful for the quantification of periodontopathogens.
