NEDD9 overexpression: Prognostic and guidance value in acute myeloid leukaemia

Abstract: Acute myeloid leukaemia (AML) is a blood cancer characterized by clonal myeloid precursors in bone marrow (BM), leading to haematopoiesis failure. 1 Clinical outcome of AML is highly heterogeneous, survive time from days to cure. 1 Cytogenetic abnormalities and gene mutations obtained at the diagnosis time provide the most important information. 2 Recently, aberrant gene expression has also been found to be associated with prognosis in AML, such as BAALC, MN1, ERG, and WT1. 3 Therefore, identification of newly… Show more

“…Underexpression of NEDD4L was observed to be correlated with normal karyotype, FLT3 and NPM1 mutations, but negatively associated with complex karyotype and TP53 mutations. Notably, a recent study also showed that abnormal NEDD9 expression, a member of NEDD family, was highly correlated with specific French-American-British (FAB) subtypes and karyotypes as well as genetic mutations, which was similar to our results [ 37 ]. These results together disclosed that NEDD4L underexpression play a key role in CN-AML biology caused by genetic mutations.…”

“…8 Moreover, NEDD9 overexpression was related to genetic abnormalities and prognosis and might serve as a possible biomarker guiding the choice between hematopoietic stem cell transplantation and chemotherapy in patients with acute myeloid leukemia after achieving complete remission. 9 However, there is little evidence showing the direct linkage between NEDD1 overexpression and the development of DLBCL. Critically, the N6methyladenosine (m6A)-based prognostic signature has been reported to be a prognostic predictor for DLBCL, and immune microenvironment governed by m6A might be a part of the mechanism.…”

“…Neural precursor cell expressed developmentally downregulated protein 1 (NEDD1) localizes to the centrosome and mitotic spindle, and the localization of key centrosomal proteins is imperative in preventing cells against uncontrolled growth and the development of cancers, suggesting that NEDD1 might be a possible candidate for anticancer treatment 8 .
