Mycoplasma mycoidessubsp.capriandMycoplasma mycoidessubsp.mycoidesLC can be grouped into a single subspecies

Abstract: -Mycoplasma mycoides subsp. capri and Mycoplasma mycoides subsp. mycoides LC can be combined into one taxon on the basis of several contributions on both DNA sequence and protein analyses reported in the literature. Moreover, for the differentiation and identification of mycoplasmas of the "mycoides cluster", we investigated the rpoB gene, encoding the β-subunit of the RNA polymerase. A segment of 527 bp of the rpoB gene was amplified from 31 strains of ruminant mycoplasmas by PCR. The nucleotide sequences wer… Show more

“…We selected 7 housekeeping genes, which were used previously for MLST analysis in other bacteria [7], [26], [27], [28]: adenylate kinase ( adk ), guanylate kinase ( gmk ), DNA gyrase subunit B ( gyrB ), dihydrolipoamide S-acetyltransferase ( pdhC ), glucose-6-phosphate isomerase ( pgi ), recombination protein ( recA ), and DNA-directed RNA polymerase beta chain ( rpoB ). About 500 bp of each housekeeping gene were sequenced representing a total of 3816 bp per strain.…”

“…Several studies have attempted to resolve the evolutionary relationships between the members of the ‘ M. mycoides cluster’ [6], [7], [8], or to infer the evolutionary history of single members within the cluster [9], [10], [11]. However, despite the recent publication of complete genome data from single isolates belonging to the different lineages [12], a comprehensive overview of the evolutionary history of the ‘ M. mycoides cluster’ and genetic relationship between populations is still lacking.…”

“…In our characterization of mycoplasmas (Abu-Groun et al, 1994;Khan et al, 2005;Lin et al, 2008;Miles and Agbanyim, 1998;Shahram et al, 2008), we have analyzed the oxidation of ten substrates by strains of two very important pathogenic rRNA sequencing (M. Shahram, unpublished), indicates that all LC and capri strains should be classified as members of a single species: provisionally "Mycoplasma capri" or "M. mycoides subsp. capri" (Lin et al, 2008;Manso-Silvan et al, 2007;Minute 10, 2007;Monnerat et al, 1999;Pettersson et al, 1996;Vilei et al, 2006).…”

“…Universal primers uvr C-F1 5'-GATCTATTTTATTAACACTACAACG-3' and uvr C-R2 5'-TCTTTAGCTGCATGAATTTG and universal rpo B-specific primers as described by Manso-Silvan et al and Vilei et al [18,19] were used at a concentration of 400 nM in a 25 ul PCR reaction volume with 1.5 mM MgCl 2 , 250 μM each dNTP, reaction buffer and 1 U InnuTaq Hot A DNA Polymerase (Analytik Jena, Jena, Germany). The temperature-time profile on a UNO II thermocycler (Biometra, Göttingen, Germany) was 95°C for 5 min and 35 times 95°C for 30 s, 50°C for 30s and 72°C for 60 s. Amplicons were purified by the DNA Clean and Concentrator Kit (Zymo Research, Irvine, USA) and subsequently sequenced by Eurofins MWG Operon (Ebersberg, Germany) using the respective forward primers.…”
