Mangifera indicaFruit Extract Improves Memory Impairment, Cholinergic Dysfunction, and Oxidative Stress Damage in Animal Model of Mild Cognitive Impairment

Abstract: To date, the effective preventive paradigm against mild cognitive impairment (MCI) is required. Therefore, we aimed to determine whether Mangifera indica fruit extract, a substance possessing antioxidant and cognitive enhancing effects, could improve memory impairment, cholinergic dysfunction, and oxidative stress damage in animal model of mild cognitive impairment. Male Wistar rats, weighing 180–200 g, were orally given the extract at doses of 12.5, 50, and 200 mg·kg−1 BW for 2 weeks before and 1 week after t… Show more

“…impaired memory and produced cholinergic dysfunction (Terry Jr and Buccafusco, 2003;Shinagawa and Shigeta, 2014;Wattanathorn et al, 2014). Consistent with those in previous reports (Yamada et al, 1999;Tran et al, 2002), data obtained by this study show that Aβ 1-40 intracisternal injection caused behavioral deficits including impaired inhibitory…”

“…2; p < 0:05). A neurotoxic derivative of choline, AF64A significantly decreases hippocampal acetylcholine contents by producing long-term presynaptic cholinergic deficits and oxidative stress resulting in impaired learning performance in rats (Thukham-Mee and Wattanathorn, 2012;Sutalangka et al, 2013a;Wattanathorn et al, 2014). Experimental data also show that AF64A administered intracisternally caused impairment of inhibitory passive avoidance, and this impairment was then partially reversed by 100 and 300 mg/kg FA ( Fig.…”

“…The central cholinergic system is involved in the process of learning and memory, which is modulated by the neurotransmitter acetylcholine (ACh) (Micheau & Marighetto 2011). AChE, a key cholinergic enzyme regulating the level of ACh in the brain, is a reliable marker for detection of central cholinergic function (Lu et al 2010b;Kumar et al 2011;Wattanathorn et al 2014). Inhibition of AChE has been used as a therapeutic strategy for Alzheimer's disease and other types of dementia (Lle o et al 2006).…”

“…In some studies it has also been demonstrated that Vimang® exerts a neuroprotective and cognitive enhancing action for mild cognitive impairment (MCI), a prodromal phase of dementia, by increasing the activity of some scavenger enzymes such as superoxide dismutase, glutathione peroxidase, catalase and lowering malondialdehyde levels [54].…”
