In Vivo Pharmacodynamic Target Assessment of Eravacycline against Escherichia coli in a Murine Thigh Infection Model

Abstract: Eravacycline is a novel fluorocycline antibiotic with potent activity against a broad range of pathogens, including strains with tetracycline and other drug resistance phenotypes. The goal of the studies was to determine which pharmacokinetic/ pharmacodynamic (PK/PD) parameter and magnitude best correlated with efficacy in the murine thigh infection model. Six Escherichia coli isolates were utilized for the studies. MICs were determined using CLSI methods and ranged from 0.125 to 0.25 mg/liter. A neutropenic m… Show more

“…16 Results from in vivo infection models with other antibiotics of the tetracycline class demonstrate that the time plasma concentrations of drug are above the MIC or AUC/MIC is the optimal PK/PD parameter for establishing efficacy. 17–23 In a PK/PD evaluation of KBP-7072 in a murine model of pneumonia, plasma AUC/MIC values for a 2-log 10 kill were 7.2 and 31.4 for Staphylococcus aureus and Streptococcus pneumoniae at 24 hours. 24 Peak KBP-7072 concentrations ranged from 0.12 to 25.2 mcg/mL.…”

“…As observed from previous PK/PD studies, the PK/PD parameter best predictive of efficacy in ERV would be the f AUC/MIC . The evidence to support f AUC/MIC is derived from murine thigh infection models evaluating E. coli strains.…”

“…The evidence to support f AUC/MIC is derived from murine thigh infection models evaluating E. coli strains. In these studies, f AUC/MIC targets were 28 for bacteriostasis and 33 for 1‐log CFU/g‐kill end points . The mean doses necessary to achieve a static and 1‐log kill were 20.43 (Standard deviation = 6.34) mg/kg every 24 hours and 34.07 (Standard deviation = 13.37) every 24 hours, respectively.…”
