In vivonon-ionizing photoacoustic mapping of sentinel lymph nodes and bladders with ICG-enhanced carbon nanotubes

Abstract: We demonstrate the feasibility of mapping a sentinel lymph node (SLN) and urinary bladder by using modified single-walled carbon nanotubes (SWNTs) as a nonionizing photoacoustic (PA) contrast agent. To improve the PA sensitivity, indocyanine green (ICG) was conjugated with SWNTs and the optical absorption of SWNTs-ICG was enhanced by approximately four times compared to that of plain SWNTs at a concentration of 0.3 µM. In vivo PA imaging results showed that the SLN and bladder were clearly visualized due to ac… Show more

“…172,210,214 SWNT has a diameter of several nm and length of 50–300 nm and ICG was attached to the surface of the SWNT. Hence it is expected that the total particle size can remain relatively small, enabling tumor targeting by iv injection.…”

“…Due to the relatively low sensitivity and poor spatial resolution of scintigraphy and single photon emission computed tomography, it is highly desirable to develop new imaging probes for other imaging modalities. The objective is to improve the detection of SLNs for either noninvasive visualization or intrasurgical guidance (10)(11)(12)(13)(14).…”

“…The limited tissue penetration of light is less critical because of the open field of view during surgery (16). For example, NIR fluorescence dyes, such as indocyanine green, have been investigated for sentinel node navigation during surgery either alone or in combination with nanoformulations (13,14). Owing to the nanometer-scale size, stability, and strong fluorescence, various nanoparticles and nanoformulations have been applied for SLN imaging and showed promising results in preclinical models (10)(11)(12).…”

“…When compared with blood in phantom studies, SWNTs show promising properties for the significant signal enhancement of for PA imaging at about 1064 nm wavelength. 290 Recently, noninvasive SWNT-enhanced PA imaging in a small animal model was applied in many biomedical applications, such as SLNs imaging, 271 cancer targeting. 290 293 The SLNs PA imaging were successfully imaged in vivo with intrinsic optical contrast at micrometer level of resolution.…”
