In Vivo Magnetic Resonance Imaging of Orthotopic Prostate Cancer

Abstract: Methods for imaging orthotopic prostate tumors within the prostate or small tumors with extension outside the prostate are needed to more closely model human prostate tumors, which are most commonly located within the gland or may extend just through the gland. By comparing MR sequences, we found that the T2-based Dixon ‘water only’ sequence best visualized tumors within the prostate of mouse models in both young and old mice and that tumor weight derived from this sequence correlated highly with ex vivo tumor… Show more

“…Ultrasound is an optimal imaging modality to monitor orthotopic prostate tumor growth ( 20 ). The advantages of ultrasound compared to MRI and bioluminescence include low cost, convenience and real time imaging, and ultrasound allows the clear delineation of orthotopic tumors from the surrounding tissues ( 12 , 21 ). CEUS has been applied in the imaging of tumors in orthotopic prostate cancer xenograft mouse models, showing orthotopic tumors are highly vascularized ( 20 , 22 ).…”

“…Established modalities that reveal and evaluate the occurrence of BCa within murine models include bioluminescence imaging (BLI), micro-ultrasound imaging (MUI), magnetic resonance imaging (MRI), and positron emission tomography (PET), as summarized in Table 1 . These modalities have provided a better understanding and visualization of cancer development [ 28 ], early detection [ 29 ], and therapeutic efficacy [ 30 , 31 , 32 ] in cancers such as breast [ 29 , 31 , 32 ], prostate [ 33 ], colon [ 28 ], and lung [ 30 ]. BLI relies on the biochemical generation of light for tumor assessment while PET utilizes radiotracers to investigate tumor burden while also providing information on the metabolic and biochemical functions.…”
