In Vivo detection of a boron‐neutron‐capture agent in melanoma by proton observed 1H ‐ 10B double resonance

Abstract: The Cesium salt of BSSB (Cs4B24H22S2), a common boron-neutron-capture-therapy (BNCT) agent, was injected into M2R mouse melanoma xenografts, and detected in vivo by 1H-observed, 10B-edited NMR spectroscopy. The technique of spin-echo difference spectroscopy, in which a proton spin-echo is detected following the alternating presence and absence of a 10B 180 degrees pulse was used. This method provides much higher sensitivity than direct 10B NMR detection, and should thus be suitable for in vivo detection in pat… Show more

“…A simple version of this approach is the subtraction of a spin-echo signal which is modulated by the 10 B-1 H coupling (achieved by applying simultaneous pulses to the boron and proton channels) from a non-modulated echo. Such an experiment was applied in-vivo to the detection of BSSB (a dimer of BSH created by formation of an S-S bond between two BSH molecules) from a mouse melanoma tumor, 42 and subsequently analyzed theoretically in more detail, ignoring relaxation. 43 However, since 1/T 1 of the boron spins can be short compared with the scalar coupling constant (particularly in-vivo), the 10 B spinlattice relaxation has a significant influence on the 1 H echo evolution, an effect known as scalar relaxation.…”

“…Mice (n ϭ 3) with tumors grown to about 1-2 cm in size were used for MRI experiments and 10 B relaxation time measurements (7). A solution (180 l) containing 0.11 M of 10 B-enriched (Ͼ95%) BSH (Boron Biologicals, Raleigh, NC) in PBS was injected into the mice tail veins.…”

“…B magnetic resonance imaging (MRI) was proposed as a non-invasive method for the in vivo assessment of BNCT agents, but its practical utilization still remains to be proven. 50,51 The other recently developed approach is 19 F MRI of fluorine-containing BPA derivatives. 52 …”
