In vivo anxiolytic and in vitro anti-inflammatory activities of water-soluble extract (WSE) of Nigella sativa (L.) seeds

Abstract: The WSE is a highly polar, gummy and mucilaginous bioactive content of the Nigella sativa (L.) seeds. This study reports the anxiolytic and anti-inflammatory effects of WSE investigated using Elevated Plus Maze (EPM) and Hole-Board Test (HBT) in adult mice and human RBCs haemolysis inhibition and protein denaturation respectively. The oral WSE treatment (100 & 200 mg/kg b.w/day) for 72 hours has exhibited slightly better anxiolytic effect (p < 0.05) through the time span (92.33 & 93.33 s) spent in the opened a… Show more

“…These findings establish a connection between BC and interleukins in BC's anti-inflammatory activity, which we have at least partially outlined in Figure 3. Similarly, other researchers speculated that BC WSE inhibits COX2 and that BC inhibits prostaglandin synthesis through interaction with COX-1 and COX-2 [46]. Both hypotheses are supported by the results of our Ingenuity pathway analysis, as depicted in Figure 3.…”

“…The treatment of each group was as described in the experimental design section. After 30 min of dosing, each mouse was placed in the apparatus and we counted the number of head dips over 5 min and determined the latency of the first head dip [21,22]. At the end of the experiment, the mice were euthanised using diethyl ether anaesthesia.…”

“…The detailed process of extraction and yield of WSE has been discussed in our earlier report (Babar et al, 2019a). This is the preliminary study of our recent study (Babar et al, 2019b) on WSE, which was preserved at 3 ±1°C in a Falcon® tube (50 mL) wrapped with aluminium foil. It was noted that 1 µL contains ≈0.88 mg (w/v) of WSE.…”

“…As the current study is a part and preliminary experiment of our recent study (Babar et al, 2019b), thus, this study protocol was also approved by the Institutional Ethics Committee (IEC) of the Department of Pharmacy, IIUC under the same reference (Pharm/PND/138/13#2019).…”
