Black cumin (Nigella sativa) is a widely used ingredient of traditional medicine for its broad-spectrum pharmacological actions, including anti-allergic, bronchial asthma, and anti-inflammatory properties. We sought to evaluate BC extracts' efficacy for their anti-allergic and anti-inflammatory properties using a comprehensive in vitro, in vivo, and silico experimental setup. To investigate whether BC extract has anti-inflammatory, anti-allergic, and analgesic therapeutic potentials in vitro and in vivo. The activity of BC was assessed through anti-allergic activity on rat basophilic leukemia-2H3 cell line, anti-inflammatory activity on J774.1A cell line, anti-inflammatory activity by carrageenan-induced rat paw edema, analgesic activity by acetic acid-induced writhing test, and ingenuity analysis of the BC extracts in inflammation control. BC exerted potent anti-allergic activity by inhibiting antigen-induced degranulation. An anti-inflammatory effect is shown by inhibiting TNF-α pro-duction. The acetic acid-induced writhing test shown a dose-dependent reduction of writhing number following BC administration. Rat paw edema test shown the dose-dependent reduction of paw edema volume following BC administration. Ingenuity Pathway Analysis (IPA) suggested BC extracts containing ferulic acid, p-coumaric acid, kaempferol, and quercetin can inhibit inflammation. This study suggests that bioactive compounds in BC extract act as an anti-allergic and anti-inflammatory agent by regulating several downstream and upstream inflammation pathways.

Background Known colloquially as akandaphal in Bangladesh, Adenia trilobata has some traditional uses. Its leaves and stems are extracted with pure methanol (MEATL, MEATS) and fractioned by n-hexane (NFATL, NFATS). The in vivo anxiolytic activity was evaluated by elevated plus maze (EPM) testing and hole-board test (HBT), whilst the locomotor activity was examined using the open-field test (OFT) and hole-cross test (HCT) and the antidepressant activity was assessed with the forced swimming test (FST) and the tail suspension test (TST). Results Regarding the anxiolytic activity, the 400-mg/kg doses of MEATL, NFATL, MEATS and NFATS exhibited maximum percentages of entry into the open arm of 33.85%, 32.23%, 30.06% and 41.84%, respectively, compare with the diazepam (69.33%). During HBT, MEATL (400 mg/kg) and NFATL (400 mg/kg) demonstrated 51.67 ± 0.88 and 57.67 ± 3.18 instances of head-dipping relative to diazepam (64.33 ± 3.16), whilst the locomotor activity showed a dose-dependent reduction in square movements and number of hole crossings. During FST and TST, the NFATL (400 mg/kg) exhibited rates of 43.32% and 57.71% time spent immobile, whilst fluoxetine experienced rates of 54.79% and 55.74%. Conclusion Adenia trilobata could be a potential component for the treatment of neuropharmacological defects. Further study is required.

The Water-Soluble Extract (WSE) is a crude bioactive phytoconstituent of Nigella sativa (L.) seeds discovered recently. The current findings report about the thrombolytic and cytotoxic effects of WSE using human blood clot lysis and brine shrimp lethality (BSL) bioassay. The thrombolytic effect of WSE (1,666.67 µg/mL) was determined via the clot and lysate weight measurements compared to streptokinase (STK) of 30,000 IU/mL and normal saline (NS) while the cytotoxicity of WSE (44.14-2,000 µg/mL) against vincristine sulfate (VCS;3.125-100 µg/mL). WSE has shown extremely statistically significant (p<0.0001) clot lysis (90.00%) compared to NS (3.76%) whilst it was also significantly different (p<0.0063) to STK (72.41%) exhibiting LC50 of 1,795.90 µg/mL vs. VCS (39.25 µg/mL) in a dose-dependent manner. The current results suggested WSE has a potent thrombolytic effect with mild dose-dependent cytotoxicity towards brine shrimp nauplii (Artemia salina). It also suggested WSE might have enzymatic roles on thrombin, fibrin, and plasmin of blood. This pharmacological action of WSE is might be due to its antioxidant property, short-chain fatty acids and/or amino acids. Further studies are highly recommended on the enzymatic role(s) and bioactive phytoconstituents of WSE.
