In vitroevaluation of functionalized decellularized muscle scaffold forin situskeletal muscle regeneration

Abstract: Current treatment options for repairing volumetric muscle loss injury involve the use of existing host tissue like muscular flaps or grafts. However, host muscle tissue may not be available and donor site morbidity, such as functional loss and volume deficiency, is often present. In this study, we developed a biofunctionalized muscle-derived decellularized extracellular matrix scaffolding system to utilize endogenous stem/progenitor cells for in situ muscle tissue regeneration. We optimized the decellularizati… Show more

“…Decellularised scaffolds are derived from xenogeneic, allogenic or autogenic skeletal muscle tissue [24][25][26]. Once deceullularisation has removed cellular material, the remaining ECM retains the native 3D microstructure, molecular composition and growth factors that support skeletal muscle regeneration [25].…”

“…Murine C2C12 are immortalised myoblasts derived from murine satellite cells and are widely used in vitro [24,43,69,[80][81][82][83][84]. They readily proliferate and differentiate under differing serum conditions and so are a useful Fig.…”

“…FGF, PDGF, and HGF promote the activation and proliferation of myogenic progenitor cells [ 101 ]. Insulin-like growth factor (IGF-I) plays an important role in all phases of satellite cell myogenesis by controlling the migration, proliferation, and differentiation of muscle satellite/progenitor cells [ 102 ].…”
