In vitro assessment of macrophage attachment and phenotype on polymerized phospholipid bilayers

Abstract: Phosphatidyl choline (PC) based materials have been found to be resistant to non-specific protein adhesion in vitro. In this study, a PC based planar supported phospholipid bilayer composed of 1,2-bis[10-(2′,4′-hexadienoyloxy)decanoyl]-sn-glycero-3-phosphocholine (bis-SorbPC or BSPC) was generated on piranha treated silicon wafers by vesicle deposition. The bilayer was polymerized with redox initiation forming a stable 4 nm thick coating. Polymerized lipid bilayers (PLBs) were characterized and tested for unif… Show more

“…Qualitative morphological criteria are typically used to determine the activation of macrophages, including the spreading, content of granules, extent of ruffling, and finer details of cell surfaces . On both alloys, the round morphology of macrophages (circularities close to 0.9) elucidates low levels of macrophage activation, as compared with the morphologies of those stimulated ones reported in literature . Detailed comparisons in terms of cell surfaces (Fig.…”

“…The count of cells adhered to the surface of the composite film materials within the microscope’s field of view was conducted utilizing a Leica DMi8 fluorescence microscope [ 40 ]. Three repetitions of experiments were carried out for each composite film sample and the comparison sample “Seprafilm”, with the number of adhered cells in each experiment assessed across three fields of view.…”

“…[ 23 ] In addition, a cell adhesion study performed on polymerized zwitterionic SLBs consisting of Bis-SorbPC showed a high degree of cell repellence towards murine-derived macrophages despite the absence of lipid lateral mobility. [ 46 ] Clearly, signifi cant fouling was observed on gel-state SLBs and SAM PEG layers, whereas on the liquid-state SLB no fouling was observed in the case of nonspecifi c adhesion of ConA. To further confi rm that using methyl-viologen-functionalized gel-state SLBs and methyl-viologen-modifi ed SAM PEG-based layers, leads to signifi cant nonspecifi c adhesion of bacterial cells to these surface exceeding by far that observed on the methyl-viologen-functionalized liquid-state SLB, we performed bacterial adhesion tests on these three layers.…”
