INTRODUCTION:Bixa orellana L. is a plant that grows in Cuba, which is held to have medicinal properties. MATERIALS AND METHODS: An experimental study was carried out to evaluate the in vitro antimicrobial action of 50% fluid extract and 10% and 20% tinctures made of the leaves of the species Bixa orellana L. The method of macrodilution in broth was used and minimal concentration with antimicrobial activity, expressed in dilutions, was determined. Strains of international reference were used. RESULTS: The tinctures at 10% and 20% of the leaf showed bactericidal action against Staphylococcus aureus at a concentration of 1:8 and 1:32 respectively, whereas only 10% tincture showed antimicrobial activity against Escherichia coli at a concentration of 1:8, however, they were not active against Pseudomona aeruginosa and Candida albicans. As to 50% fluid extract, it only showed bactericidal activity against Staphylococcus aureus at a concentration of 1:4. CONCLUSION: Both, fluid extract at 50% and tinctures at 10% and 20% of the leaf were active against Staphylococcus aureus at lower concentrations compared to the rest of microorganisms, but they had no antimicrobial activity against Candida albicans or Pseudomona aeruginosa. 50% fluid extract showed antimicrobial activity only against Staphylococcus aureus. Tincture at 10% confirmed activity against Staphylococcus aureus and Escherichia coli. Tincture at 20%was of use only against Staphylococcus aureus. Staphylococcus aureus turned out to be the most sensitive germ to the pharmaceutical forms studied at lower concentrations. Tincture at 10% showed a wider spectrum of antimicrobial activity.

Bixa orellana leaf preparations have been used for many years by indigenous people for a variety of medicinal applications. Published research studies in animals indicate that various extracts of Bixa leaves exhibit antioxidant, broad antimicrobial (antibacterial and antifungal), anti-inflammatory, analgesic, hypoglycemic, and antidiarrheal activities. No studies have specifically assessed the ability of leaf extracts to inhibit urogenital infections although Bixa products have been used in folkloric medicine to treat gonorrhea and other infections. Few human studies have been conducted and published using Bixa leaf preparations. Many more studies have been conducted and published involving Bixa seed (annatto) extracts than with leaf extracts. No subchronic safety (toxicity) studies have been conducted in animals. A 6 month study in humans given 750 mg of leaf powder per day demonstrated no significant or serious adverse effects. Bixa leaf extracts appear to be safe when given under current conditions of use. However, additional human and animal controlled safety and efficacy studies are needed. In addition, detailed chemical analyses are required to establish structure-function relationships.

Chemical composition of pomegranate juice can vary due to cultivar, area of cultivation, ripening, climate, and other variables. This study investigates the polyphenolic composition and antioxidant activity of juices obtained from six old Italian pomegranate cultivars. Fruit accessions physicochemical characteristics were determined. Total polyphenols content (TPC), anthocyanin content (TAC) and proanthocyanidin content (TPAC) were measured in the juice samples. Phenolic bioactive molecules were analyzed by HPLC-photodiode array (PDA)/ESI-MS in all the pomegranate juices. In total, seven nonanthocyanidinic and six anthocyanidinic compounds were identified. The six anthocyanins were found in all juices although at different amounts. These results were correlated with antioxidant activity measured by three different chemical assays: 2,2 diphenyl-1-picrylhydrazyl (DPPH(•) ) scavenging activity assay, Trolox equivalent antioxidant capacity (TEAC) method and ferric reducing-antioxidant power (FRAP) assay. Pomegranate juices obtained by six different varieties show variable polyphenolic content and antioxidant activity. The antioxidant capacity methods used have shown variable sensitivity, supporting the hypothesis that different methods for the assessment of antioxidant capacity of food compounds are indeed necessary, due to complexity of sample composition and assay chemical mechanism and sensitivity. Juices from Italian pomegranate show good levels of polyphenols content and antioxidant activity making them potential candidates for employment in the food industry.
