In Vitro and In Vivo Inhibition of Skin Matrix Metalloproteinases by Pothomorphe umbellata Root Extract

Abstract: Exposure to UV radiation up-regulates the synthesis of matrix metalloproteinases (MMPs), a group of matrix-degrading enzymes. MMPs are regarded as promising therapeutic targets and the development of effective inhibitors is an important research focus. The plant Pothomorphe umbellata has been shown to exert a potent antioxidant activity on the skin and to delay the onset and reduce the incidence of UVB-induced chronic skin damage. The aim of the present study was to determine the effect of P. umbellata ethanol… Show more

“…The organic phase was separated, and the aqueous phase was extracted with ethyl acetate (3ϫ30 ml). The combined extracts were washed with saturated brine (30 ml) and dried with MgSO 4 . Removal of ethyl acetate solvents on a rotary evaporator followed by vacuum drying yielded 91% and 94% of 7a and 7b, respectively.…”

“…Oxidative stress is thought to play a central role in initiating and driving the signaling events that lead to the cell response following exposure to UV irradiation. 4,5) Exposure of human skin to UV radiation causes numerous biological effects in the skin, including premature aging characterized by wrinkles, leathery texture and mottled pigmentation. 6,7) The mechanism of skin wrinkles is primarily due to the loss of macromolecules making up the dermal matrix, among which collagen is the major component.…”

“…These results suggest that SBFE may contribute to a recovery from UV-induced cell death through the regulation of apoptotic protein expression and that it may have potential therapeutic utility in ameliorating UV-induced skin ageing. 한편, UV조사로 세포 내에 유도된 산화적 스트레스는 세포 반응을 개시하고 일련의 반응을 유도하는 과정에서 매우 중요 한 역할을 한다 [25,30]. 특히, 세포 내 ROS를 생성시켜 미토콘 드리아를 비롯한 세포의 기능을 손상시킨다 [6].…”
