Summary Intraperitoneal injection of (BALB/c X C57BL/6) Fl mice with live, but not killed Salmonella enteritidis 1 lRX (1 lRX) induced T cells in the spleen and peritoneal cavity which were able to proliferate in vitro in response to two different forms of 1 lRX antigens. The majority of cells which proliferated were L3T4+ T cells and most of the response was restricted by the I-A locus of the H-2 major histocompatibility complex, although a smaller K region restricted response was also detected. T cells able to respond to 1 lRX antigens could only be demonstrated when non-adherent lymphoid cell suspensions from immunized mice were used, and usually a limited response was obtained unless smalt numbers of adherent cells present in normal peritoneal cell suspensions were added. Cells cultured in vitro for 3 days were able to mediate local transfer of delayed type hypersensitivity and secondary immunization did not enhance the reactivity of responding cells to 11RX antigens.

Summary The B16 melanoma of C57BL/6 mice immunizes very poorly, even against its own major histocompatibility complex (MHC) antigens. B16 cells expressed both H‐2K and H‐2D antigens in vitro as judged by binding of monoclonal antibodies to these antigens in indirect immunofluorescence staining. The in vivo MHC antigen expression of B16 was examined and compared with that of a second C57BL/6 tumour, the Lewis lung carcinoma (3LL). whose defective immunogenicity has been attributed to a selective deficiency‐ in H‐2K antigen expression. We found that 125I‐labeiled cells of both tumours expressed sufficient allo‐antigen in vivo to be lysed in BALB/c mice which had been pre‐immunized with C57BL/6 lymphoid cells. 125I‐B16 cells were also lysed in MHC‐recombinant mice which had been immunized against either H‐2Kb or H‐2Db, indicating that B16 cells express both of these MHC antigens in vivo. This contrasted with our findings with 125I‐3LL cells which were destroyed in mice immunized against H‐2Db but not in those immunized against H‐2Kb. Thus, B16 illustrates a different deficiency in tumour cell immunogenicity which appears not to be attributable to an absence of either of the class 1 MHC antigens.

Summary In an attempt to characterize the effector cells responsible for tumour resistance in Salmonella enteritidis 11RX‐immunized mice the anti‐macrophage agent trypan blue was used in both in vivo and in vitro experiments. Resistance was measured in vivo by the clearance of 125I from she peritoneal cavity of mice injected intraperitoneally with 125I‐5‐iododeoxyuridine labelled Ehrlich Ascites Tumour (EAT) cells. The in vitro correlate was measured by lysis of 51Cr‐labelled tumour cells by peritoneal cells (PC) from 11RX‐immunized mice. Pre‐treatment of resistant mice with trypan blue greatly reduced both 125I clearance and 51Cr release. The in vitro cytolytic activity was non‐specific. Fractionation of cytotoxic PC on the basis of adherence to plastic or nylon wool and buoyant density, coupled with the use of appropriate cell targets, showed that the bulk of cytotoxic activity resided with macrophages, with some contribution from other cells such as natural killer cells. Killing of labelled tumour cells could be inhibited by competition with unlabelled cells or by separating the PC and tumour cells by a cell impermeable membrane. This showed that close association between the effector and target cells was necessary before killing could occur.
