Gleditsia sinensis thorn extract inhibits human colon cancer cells: the role of ERK1/2, G2/M‐phase cell cycle arrest and p53 expression

Abstract: The thorns of Gleditsia sinensis are used as a medicinal herb in China and Korea. However, the mechanisms responsible for the antitumor effects of the water extract of Gleditsia sinensis thorns (WEGS) remain unknown. HCT116 cells treated with the WEGS at a dose of 800 μg/mL (IC₅₀) showed a significant decrease in cell growth and an increase in cell cycle arrest during the G2/M-phase. G2/M-phase arrest was correlated with increased p53 levels and down-regulation of the check-point proteins, cyclinB1, Cdc2 and C… Show more

“…ERK, JNK and p38 MAPK constitute 3 major subfamilies of MAPKs that appear to mediate cellular responses, including proliferation, differentiation and apoptosis (30). Gleditsia sinensis thorns (WEGS) induced phosphorylation of ERK1/2, p38 MAPK and JNK in human colon cancer cells (31). Danshensu increased phosphorylation of Akt and ERK1/2 in H9c2 cardiomyocytes (32).…”

“…Growth inhibition caused by ERK1/2 loss is rescued by reintroducing ERK2, but not by activating ERKs effectors that promote G1 cell cycle progression (38). Some natural compounds, such as oridonin (39) and Gleditsis sinensis thorn extract, reportedly induce G2/M arrest through ERK signaling (40). Our data showed that induction of cell cycle arrest by norathyriol may be mediated through ERKs signaling because norathyriol strongly suppresses ERK1 and 2 phosphorylation and kinase activities.…”

“…However, adverse effects of bevacizumab have been reported, such as hypertension, and there are a number of potential problems that warrant caution in human clinical trials [3,45]. Recently, several plant-derived compounds have been shown to be potent inhibitors of angiogenesis both in vitro and vivo [19,29,50]. Therefore, herbal medicine with few side effects is a promising strategy for anti-angiogenic tumor therapy.…”
