Ex Vivo Bone Formation in Bovine Trabecular Bone Cultured in a Dynamic 3D Bioreactor Is Enhanced by Compressive Mechanical Strain

Abstract: Our aim was to test cell and trabecular responses to mechanical loading in vitro in a tissue bone explant culture model. The ZetOS TM system provides the ability to exert cyclic compression on cancellous bone cylinders (bovine sternum) cultured in forced flow circumfusion chambers, and allows assessing mechanical parameters of the cultivated samples. We evaluated bone cellular parameters through osteocyte viability test, gene and protein expression and histomorphometric bone formation rate, in non-loaded versu… Show more

“…Consistently, dynamic compression enhances the vasculogenic activities of circulating endothelial precursors (EPCs) seeded in the demineralized bone matrix scaffolds (Kong et al, 2012). The relationship between mechanical inputs and resulting biological tissue structure, composition, and metabolism is reported and discussed in several other papers and reviews (David et al, 2008;Desmoulin et al, 2013;Gardel, Serra, Reis, & Gomes, 2014;Gaspar, Gomide, & Monteiro;S.-T. Li et al, 2014;van Griensven et al, 2009).…”

“…Our original culture chamber was developed to be compatible with the Zetos ® compression device (David et al, 2008;Davies et al, 2006;Dumas et al, 2009) which G Bouet et al In vitro 3D bone culture model can exert cyclic compression on a mobile piston through a piezo-electric actuator controlled by a computer program (Fig. 1D).…”

“…Chick limb organ culture in the 21 st century perfusion of nutrients throughout the tissue (Bilodeau and Mantovani, 2006;Butler et al, 2000;David et al, 2008;El Haj and Cartmell, 2010;Mauney et al, 2004;Yu et al, 2004). A wide variety of bioreactor types exist that include rotating oxygen-diffusing vessels, filled with culture media, and perfusion systems incorporating controlled flow back and forth within the construct.…”
