DGAT1 promoter polymorphism associated with alterations in body mass index, high density lipoprotein levels and blood pressure in Turkish women

Abstract: Triglyceride synthesis is catalyzed by acyl CoA:diacylglycerol acyltransferases (DGAT), microsomal enzymes that use diacylglycerol and fatty acyl CoAs as substrates. Because DGAT1 expression is up-regulated during adipocyte differentiation and DGAT1 deficiency is associated with leanness in mice, we hypothesized that alterations in DGAT1 expression may affect human body weight. We identified five polymorphisms in the human DGAT1 promoter and 5' non-coding sequence in a random Turkish population. Functional ana… Show more

“…The DGAT T79C C allele is associated with increased promoter activity and higher BMI in Turkish women. 51 However, this has not been confirmed in a large French cohort. 52 Other genes involved in TAG storage such as lipoprotein lipase (LPL), acylation stimulating protein (ASP) and fatty acid binding proteins (FABPs) are not implicated in susceptibility to obesity.…”

“…Thus, transcription of the MGAT2 gene may similarly be subject to transactivation by PPAR␣ or other isoforms of PPAR in response to elevated levels of fatty acids in the mucosa to increase the rate of dietary absorption. Future studies are also needed to depict a role of MGAT2 in human obesity, such as a possible linkage of MGAT2 polymorphism with certain types of human obesity as demonstrated with DGAT (47,48). Furthermore, generation and characterization of mice deficient in MGAT2 expression will shed important light on a role for MGAT2 in dietary fat absorption, as well as in diet-induced obesity.…”

“…These findings indicate a robust upregulation of DGAT1 and DGAT2 during adipogenesis and suggest the involvement of C/EBPa or PPARg, transcription factors that control the expression of many genes in adipogenesis. A PPAR binding site occurs in the DGAT1 promoter (89), and thiazolidinediones, which activate PPARg, increase DGAT1 mRNA levels in adipocyte cell lines and adipose tissues of mice and humans (90). C/ EBPb and C/EBPa increase the expression of DGAT2 mRNA in adipogenesis (91).…”

“…Several common sequence polymorphisms are present in the 5′ noncoding sequences of the DGAT1 gene (89). One of these polymorphisms, C79T, affects promoter activity in cultured cells and has been associated with alterations in body mass index, diastolic blood pressure, and HDL cholesterol levels in Turkish women (89).…”
