Cstf2t Regulates expression of histones and histone‐like proteins in male germ cells

Abstract: Formation of the 3' ends of mature mRNAs requires recognition of the correct site within the last exon, cleavage of the nascent pre-mRNA, and, for most mRNAs, addition of a poly(A) tail. Several factors are involved in recognition of the correct 3'-end site. The cleavage stimulation factor (CstF) has three subunits, CstF-50 (gene symbol Cstf1), CstF-64 (Cstf2), and CstF-77 (Cstf3). Of these, CstF-64 is the RNA-binding subunit that interacts with the pre-mRNA downstream of the cleavage site. In male germ cells … Show more

“…CstF64/tau is encoded by Cstf2t, a paralog of the Cstf2 gene that encodes CstF64. Cstf2t is expressed in germ cells, also has a role on polyadenylation, being indispensable for spermatogenesis in rats (Harris et al, 2016;Grozdanov et al, 2018a). CstF64/tau binds to sequences rich in U>G also downstream of the PAS (Mandel et al, 2008).…”

“…Our laboratory has examined further roles for τCstF‐64 in global mRNA expression (Li et al, ), APA and splicing of the Crem mRNA (Grozdanov, Amatullah, Graber, & MacDonald, ), and expression of histone mRNAs (Grozdanov, Li, Yu, Yan, & MacDonald, ) in testis. We also found important roles for τCstF‐64 in embryonic stem cells (Youngblood et al, ) and during endoderm differentiation (Youngblood & MacDonald, ).…”

“…For example, the gene Cleavage Stimulation Factor Subunit 2 Tau Variant ( Cstf2t ) has been demonstrated to be an important stage-specific regulator of Crem mRNA processing that controls Crem polyadenylation in mouse testis. Cstf2t can lead to an overall decrease of the Crem mRNAs generated from internal promoters in Cstf2t −/− mice ( 30 , 31 ). Therefore, these 140 RBPs can be promising candidates for RNA-Seq studies in the future.…”
