Clinanthus microstephium, an Amaryllidaceae Species with Cholinesterase Inhibitor Alkaloids: Structure−Activity Analysis of Haemanthamine Skeleton Derivatives

Abstract: Plants of the Amaryllidaceae family are well-known (not only) for their ornamental value but also for the alkaloids they produce. In this report, the first phytochemical study of Clinanthus genus was carried out. The chemical composition of alkaloid fractions from Clinanthus microstephus was analyzed by GC-MS and NMR. Seven known compounds belonging to three structural types of Amaryllidaceae alkaloids were identified. An epimeric mixture of a haemanthamine-type compound (6-hydroxymaritidine) was tested as an … Show more

“…Meerow & A. Cano and Clinanthus incarnatus (Kunth) Meerow has recently been reported [ 14 ]: lycorine- and haemenanthamine/crinine-type alkaloids were found in both species, together with galantamine-type alkaloids in C. incarnatus [ 14 ]. Lycorine and haemanthamine-type alkaloids were also reported in Clinanthus microstephium , as well as a homolycorine-type alkaloid [ 15 ].…”

“…A molecular docking study was performed on 6-hydrohymaritidine by Adessi et al (Adessi et al 2019). The b:a epimeric mixture of 6-hydroxymaritidine, isolated from Hippeastrum reticulatum that showed the highest activity within the studied seven haemanthamine skeleton type alkaloids, was subject to deep pre-and post-docking analysis, including a detailed consideration of the ligand protonation state at physiological pH, and ab-initio DFT calculations for estimation the conformers ratio in water.…”

“…Although chemical and pharmacological interest in the genus Clinanthus has increased in recent years, only seven published articles about this genus can be found in the literature, and five of them correspond to the description of new species [ 1 , 9 , 10 , 21 , 22 ]. The first report of a chemical characterization and bioactivity evaluation of a species from this genus was carried out by Adessi and co-workers [ 23 ], who reported the inhibition of AChE and BChE enzymes by the alkaloid 6-hydroxymaritidine, extracted and purified from C. microstephium [ 23 ]. Another study from 2021 reported the chemical characterization and the antiplasmodial activity of two Clinanthus species ( C. incarnatus and C. ruber ) [ 24 ].…”
