Citrus limonextract: possible inhibitory mechanisms affecting testicular functions and fertility in male mice

Abstract: The effect of oral administration of 50% ethanolic leaf extract of Citrus limon (500 and 1,000 mg/kg body weight/day) for 35 days on fertility and various male reproductive endpoints was evaluated in Parkes strain of mice. Testicular indices such as histology, 3β- and 17β-HSD enzymes activity, immunoblot expression of StAR and P450scc, and germ cell apoptosis by TUNEL and CASP- 3 expression were assessed. Motility, viability, and number of spermatozoa in the cauda epididymidis, level of serum testosterone, fer… Show more

“…The affected tubules in testes of mice after FSE treatment exhibited intraepithelial vacuolation, germ cell exfoliation, giant cell formation, degeneration of germ cells, and marginal condensation of chromatin in round spermatids. Such changes have also been shown in mice testes after treatment with Allamanda cathartica (Singh & Singh, 2008), Dalbergia sissoo (Verma & Singh, 2014) and Citrus limon (Singh & Singh, 2016). The results also showed a significant decrease in testis weight after FSE treatment, and a decrease in testicular weight is considered as an indication of morphological abnormalities that may have resulted from the FSE treatment (Grima et al, 2001).…”

“…This is not surprising because there appears absence of good correlation between serum testosterone levels and the sexual function as symptoms of hypogonadism have been noted in men even in the presence of normal testosterone concentrations (Travison et al ., ). Further, it is relevant to mention that there are also reports of marked decrease in serum level of testosterone with no effect on libido in P mice, for example, after treatment with aqueous leaf extract of Dalbergia sissoo (Verma & Singh, ) and 50% ethanolic leaf extract of Citrus limon (Singh & Singh, ). The absence of any alterations in the weights of the liver, spleen, kidney and adrenal glands, serum levels of ALT, AST and creatinine, and in haematological parameters in Mimusops‐treated mice suggests that the treatment does not produce systemic toxic effects.…”

“…The negative control slides were processed in a similar way without using TdT in TUNEL reaction solution. TUNEL‐positive apoptotic germ cells were counted in 20 tubular cross sections per testis from each animal in a group, and mean value of apoptotic cells in each group was presented (Singh & Singh, ).…”
