Candida Administration Worsens Uremia-Induced Gut Leakage in Bilateral Nephrectomy Mice, an Impact of Gut Fungi and Organismal Molecules in Uremia

Abstract: The impact of gut fungi and (1→3)-β-d-glucan (BG), a major fungal cell wall component, on uremia was explored by Candida albicans oral administration in bilateral nephrectomy (BiNx) mice because of the prominence of C. albicans in the human intestine but not in mice. As such, BiNx with Candida administration (BiNx-Candida) enhanced intestinal injury (colon cytokines and apoptosis), gut leakage (fluorescein isothiocyanate [FITC]-dextran assay, endotoxemia, serum BG, and bacteremia), systemic inflammation, and l… Show more

“…drugs) [47,60] in FcgRIIb -/-mice, due to the absence of the inhibitory signaling [29,43], are previously mentioned. Here, a single stimulation of Ahr activation alone did not induce inflammatory responses, in either FcgRIIb -/-or WT macrophages, but LPS alone increased Ahr expression in macrophages from both mouse strains.…”

“…The hyper-immune responsiveness toward the organismal elements (pneumococcal antigens and LPS) [38,43] and the non-infection related components (uremic toxin and drugs) [47,60] in FcgRIIb −/− mice, due to the absence of the inhibitory signaling [29,43], are previously mentioned. “…Perhaps, NSAIDs induced uremia and uremia-induced inflammation exacerbates lupus activity [34,35]. However, NSAIDs nephropathy might not be a main lupus exacerbating factor in NSAIDs-mouse model because (i) the uremia in NSAIDs model (no obvious renal histological damage) is less severe than other direct renal damage models (ischemia and bilateral nephrectomy) [61,62] and (ii) endotoxemia from uremia (an indirect mucosal damage) is less severe than the endotoxemia from the direct gut mucosal injury caused by NSAIDs [63,64],…”

“…Additionally, macrophages are the heterogeneous immune cells with pleiotropic functions; the pro- and anti-inflammatory properties [ 29 , 30 , 31 ], which use glycolysis and mitochondria, respectively, as the main metabolic signatures [ 32 , 33 , 34 ]. Interestingly, the cell energy status of macrophages could be altered by either PAMPs, including endotoxin [ 35 ], or damage associated molecular patterns (DAMPs), including uremic toxins [ 36 ]. Although the exacerbation of macrophage inflammatory responses after activation by uremic toxins and endotoxin in synergy [ 36 ] through the cross-talk between TLR-4 and Dectin-1 [ 37 , 38 , 39 ] is previously demonstrated, an impact of fungal molecules on macrophage energy status is still interesting.…”
