Burkholderia anthinasp. nov. andBurkholderia pyrrocinia, two additionalBurkholderia cepaciacomplex bacteria, may confound results of new molecular diagnostic tools

Abstract: Nineteen Burkholderia cepacia-like isolates of human and environmental origin could not be assigned to one of the seven currently established genomovars using recently developed molecular diagnostic tools for B. cepacia complex bacteria. Various genotypic and phenotypic characteristics were examined. The results of this polyphasic study allowed classification of the 19 isolates as an eighth B. cepacia complex genomovar (Burkholderia anthina sp. nov.) and to design tools for its identification in the diagnostic… Show more

“…Table 1 shows that strain MSh1 T can be differentiated biochemically from the closely related members of the genus Burkholderia . It was shown that strain MSh1 T differs from the other Bcc species by the ability to acidify adonitol, having arginine dihydrolase activity and inability to assimilate N-acetyl-glucosamine (Vandamme et al, 1997, 2000, 2002; Coenye et al, 2001; Henry et al, 2001; Vanlaere et al, 2008, 2009; Peeters et al, 2013). …”

“…Data for the other Bcc species were obtained from Coenye et al (2001), Henry et al (2001), Peeters et al (2013), Vandamme et al (2002), Vandamme et al (1997), Vandamme et al (2000), Vanlaere et al (2009), and Vanlaere et al (2008). “…This is the first report of B. cepacia on onion in Japan. The B. cepacia complex has been subdivided by phenotypic markers and DNA-DNA relatedness into at least nine genomic species or genomovars (Coenye et al 2001), including B. cepacia sensu stricto (genomovar I), B. multivorans (genomovar II), B. cenocepacia (genomovar III) , B. stabilis (genomovar IV), B. vietnamiensis (genomovar V), B. dolosa (genomovar VI) (Biddik et al 2003;Vermis et al 2003), B. ambifaria (genomovar VII), B. anthina (genomovar VIII) (Vandamme et al 2002), and B. pyrrocinia (genomovar IX) (Vandamme et al 2002). Most of the B. cepacia isolates from plants were assigned to genomovar I or III (Balandreau et al 2001;Lee and Shiao 2003), but there is no clear-cut differential biochemical test to distinguish genomovar III from genomovar I.…”

“…The RFLP patterns were manually analysed and compared to those shown by Bcc reference strains (Mahenthiralingam et al 2000). Also, the putative species status of each Bcc isolate was determined by species-specific PCR tests, according to the procedures described previously (Mahenthiralingam et al 2000;Vandamme et al 2002;Pirone et al 2005). Further resolution of the correct species was achieved with nucleotide sequence analysis of the Bcc recA gene.…”

“…Phylogenetic analysis was performed on 6 novel recA sequences and 38 previously published Bcc recA gene (Mahenthiralingam et al 2000;Vandamme et al 2002;Coenye and Vandamme 2003;Baldwin et al 2005;Payne et al 2005;Zhang and Xie 2007). Nucleotides of the recA gene were aligned using CLUSTAL W. Phylogenetic and molecular evolutionary analyses were conducted using the genetic distance-based neighbor-joining algorithms within MEGA version 4.0 (http://www.megasoftware.net/).…”
