bla VIM-2 Cassette-Containing Novel Integrons in Metallo-β-Lactamase-Producing Pseudomonas aeruginosa and Pseudomonas putida Isolates Disseminated in a Korean Hospital

Abstract: We investigated the phenotypic and genetic properties of metallo-␤-lactamase-producing Pseudomonas isolates collected at a tertiary-care hospital in Korea since 1995. The prevalence of imipenem resistance among Pseudomonas aeruginosa isolates reached 16% in 1997, when 9% of the resistant organisms were found to produce VIM-2 ␤-lactamase, a class B enzyme previously found only in P. aeruginosa isolates from Europe. VIM-2-producing isolates of Pseudomonas putida were also detected. Resistance was transferable fr… Show more

“…The PCR analysis for the MBL genes showed predominantly VIM type (43%) genes and IMP (14%) type in our setting, similar prevalence was seen ranging from 18 % to 56% for VIM and 22 % to 48% for IMP type in other studies (Doosti et al, 2013;Irfan et al, 2008;Kim et al, 2012;Lee et al, 2002). In another study, there was prevalence of 17.8% VIM type MBLs in the Citrobacter spp (Praharaj et al, 2016).…”

“…While 5' conserved region of class 1 integron includes class 1 integrase gene encoding a site-specific integrase (intI) and recombination site, 3' conserved region includes sulI and qacEΔ1 genes [18]. Class 1 integrons have been reported in P. aeruginosa isolates [8,[19][20][21][22][23][24][25][26][27]. Organization of class 2 integrons is similar to the class 1 integrons and they are inserted in the Tn7 transposons family.…”

“…DNA extraction was carried out by boiling (15). Briefly, a loopful of an overnight grown culture of each test isolate was suspended in 500 µL of sterile double-distilled water, boiled at 100°C for 10 minutes, and centrifuged at 13,000 × g for 10 minutes.…”
