Arabidopsis CLV3 Peptide Directly Binds CLV1 Ectodomain

Abstract: CLV1, which encodes a leucine-rich repeat receptor kinase, and CLV3, which encodes a secreted peptide, function in the same genetic pathway to maintain stem cell populations in Arabidopsis shoot apical meristem. Here, we show biochemical evidence, by ligand binding assay and photoaffinity labeling, that the CLV3 peptide directly binds the CLV1 ectodomain with a dissociation constant of 17.5 nM. The CLV1 ectodomain also interacts with the structurally related CLE peptides, with distinct affinities depending on … Show more

“…Intriguingly, the RxR motif is largely conserved in this subfamily of LRR-RKs ( Figure 1B). Furthermore, several members of this LRR-RK subfamily have been shown to recognize small peptides ending with histidine or asparagine [20][21][22][23][24][25]. These findings prompted us to hypothesize that members of the LRR XI subfamily may act as receptors of small peptides with a free C-terminal histidine or asparagine.…”

“…To prepare HAR1 L246F -HT, the amino-acid substitution L246F was introduced into HAR1-HT by PCR-based site-directed mutagenesis. Transformation of tobacco BY-2 cells and preparation of microsomal fractions were described previously 36 . To confirm expression, HaloTag-fused proteins were specifically labelled using HaloTag TMR (Promega), separated by SDSpolyacrylamide gel electrophoresis, and visualized by a fluorescent image analyzer with a 523 nm excitation filter and a 580 nm emission filter.…”

“…The CLAVATA3 (CLV3)/EMBRYO SURROUNDING REGIONrelated (CLE) family of signalling peptides plays crucial roles in the regulation of stem cell maintenance in the meristems 3 . In the shoot apical meristem of Arabidopsis thaliana, CLV3 peptide secreted from stem cells restricts the expression domain of the WUSCHEL (WUS) homeobox transcription factor gene to the organizing centre via three receptor pathways, CLV1, CLV2-CORYNE (CRN) and RECEPTOR-LIKE PROTEIN KINASE 2 (RPK2) [4][5][6][7][8][9][10] . CLV3-WUS signalling makes a negative feedback loop to maintain the homoeostasis of stem cell population in the shoot apical meristem [11][12][13] .…”
