ANGPTL1 Interacts with Integrin α1β1 to Suppress HCC Angiogenesis and Metastasis by Inhibiting JAK2/STAT3 Signaling

Abstract: Downregulation of tumor suppressor signaling plays an important role in the pathogenesis of hepatocellular carcinoma (HCC). Here, we report that downregulation of the angiopoietin-like protein is associated with vascular invasion, tumor thrombus, metastasis, and poor prognosis in HCC. Ectopic expression of in HCC cells effectively decreased their and tumorigenicity, cell motility, and angiogenesis. shRNA-mediated depletion of exerted opposing effects. promoted apoptosis via inhibition of the STAT3/Bcl-2-mediat… Show more

“…Then mesenchymal-like cells invade into vessels where some of them become anoikis resistant and translocate to distant organ to form metastases. Particularly, the metastasis of HCC shares considerable similarities with other cancer types, whereas EMT program and enhanced angiogenesis have been addressed as critical events driving HCC metastasis [145,146].…”

“…DADS can inhibit the CCL2 (CC chemokine-2) releasing induced by TNF-α to play an antiangiogenesis role in MDA-MB-231 cells. The decreasing level of CCL2 attenuates the phosphorylation of ERK which is a key molecule of the MAPK signaling pathway, leading to the weakening of angiogenesis in tumor cells [111]. Activation of ERK and Akt signaling can promote the angiogenesis of HCC (hepatocellular carcinoma) [112].…”

“…STAT3 and HIF‐1α, two major transcription factors that regulate VEGF, have been found to be consistently upregulated in various cancers and associated with poor clinical outcomes in patients . STAT3 is a well‐characterized oncogene that affects various biological processes including promoting cell proliferation and survival by regulating Cyclin D1 and Bcl‐2, inducing tumor invasion and metastasis by regulating E‐cadherin, MMP‐9, promoting angiogenesis by targeting VEGF and HIF1α expression . Angiogenesis is also reported to be linked with STAT3 .…”
