<i>Alkaloids of Genista burdurensis</i>

In the current study, a number of alkaloids including retamine, cytisine, and sparteine (quinolizidine-type), yohimbine and vincamine (indole-type), scopolamine and atropine (tropane-type), colchicine (tropolone-type), allantoin (imidazolidine-type), trigonelline (pyridine-type) as well as octopamine, synephrine, and capsaicin (exocyclic amine-type) were tested in vitro for their inhibitory activity against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) at 1 mg/ml concentration by the Ellman method using an ELISA microplate reader. Among the alkaloids tested, only capsaicin exerted a remarkable inhibitory effect towards both AChE and BChE [(62.7 ð 0.79)% and (75.3 ð 0.98)%, respectively]. While the rest of the alkaloids did not show any significant inhibition against AChE, three of the alkaloids, namely retamine, sparteine, and yohimbine, exerted a noteworthy anti-BChE effect as compared to galanthamine, the reference drug.

at ô = 3.60 ppm due to COOCH3 group. From its 1H-NMR, '3C-NMR, and mass spectrum, this compound was identified as the methyl ester of compound 2, which could be an artifact formed during the extraction procedure. The compound 4 showed the molecular ion peak at mlz = 412 (C29H480). This compound was identified as stigmasterol from the comparison of its 'H-NMR and '3C-NMR data with those of reported data (4, 5). The mass spectrum of compound 5 indicated the molecular ion peak at m/z = 416 corresponding to the molecular formula C29H520. This compound was identified as 3-stigmastanol on the basis of a 1H-NMR comparison (4). Materials and Methods4 11/2 kg wet wt of Botryocladia leptopoda was collected in April 1985 around Patcha near Karachi and soaked in MeOH for a week. The MeOH ectract after evaporation under reduced pressure yielded 5.8 g of crude extract, which was partitioned between ethyl acetate and H20. The crude ethyl acetate thus obtained (1.5 g) after evaporation was subjected to column chromatgraphy using hexane, hexane : ether, ether, chloroform: MeOH, and finally with pure MeOH. The eluate obtained from hexane: ether (90: 10), (75 :25), and (70 : 30) yielded compounds 1,2 and 3, re-The 'H-NMR and 13C-NMR spectra were recorded in CDCI3 on Bruker AM-300 at 300 MHz and 50.15 MHz, respectively.
