Hypoxic Induction of the Regulator of G-Protein Signalling 4 Gene Is Mediated by the Hypoxia-Inducible Factor Pathway

Abstract: The transcriptional response to hypoxia is largely dependent on the Hypoxia Inducible Factors (HIF-1 and HIF-2) in mammalian cells. Many target genes have been characterised for these heterodimeric transcription factors, yet there is evidence that the full range of HIF-regulated genes has not yet been described. We constructed a TetON overexpression system in the rat pheochromocytoma PC-12 cell line to search for novel HIF and hypoxia responsive genes. The Rgs4 gene encodes the Regulator of G-Protein Signallin… Show more

“…We have recently found, for example, that hyperthermia (heat shock) markedly alters the expression of several SZ, BD and ASD candidate genes, including ZNF804A , ARNT2 and SMARCA2 , in a model of the human fetal telencephalon derived from iPSCs [ 66 , 96 ]. In addition, in mouse models, gene x environment interactions involving the SZ, BD, and ASD candidate genes DISC1 , NRG1 , RGS4 , and AKT1 have been described [ 97 102 ].…”

“…In this in vitro ischemia model, the growth medium of the cells was replaced with medium lacking glucose (HBSS Phenol red medium) and placed in a chamber that was rendered anaerobic by a sachet containing ascorbic acid (AnaeroGenTM, OXOID, Germany) [10]. Resazurin, an anaerobic indicator (OXOID, Germany) sensitive to changes in oxygen levels, was placed in the chamber and the lid of the chamber was tightly closed and placed in the incubator at 37°C.…”

“…The abundance of EPO mRNA was about 20-fold higher than in total kidneys of nontreated mice. In parallel, also the expression levels of other genes supposed to be triggered by HIF-2, such as adrenomedullin (ADM), cyclooxygenase-2 (COX-2), regulators of G-protein signaling (RGS) 2, 4, and 5, or PHD3, [32][33][34][35][36][37][38][39] were clearly increased. In glomeruli of mice Enzymes such as enolase 1 and lactate dehydrogenase 1 were also increased in Vhl-deficient glomeruli and remained elevated in glomeruli with an additional knockout of HIF-2α, suggesting that they were regulated by HIF-1.…”
