Hypoxic culture conditions induce increased metabolic rate and collagen gene expression in ACL‐derived cells

Abstract: There has been substantial effort directed toward the application of bone marrow and adipose-derived mesenchymal stromal cells (MSCs) neg , CD45 neg )-and describe their proliferative and differentiation potential, collagen gene expression and metabolism in both normoxic and hypoxic environments, and their trophic potential in vitro. All three groups of cells (LIC, LPC, and LFF) isolated from adult human ACL exhibited progenitor cell characteristics with regard to proliferation and differentiation potential … Show more

“…High donor variability for human BMSCs was demonstrated previously, suggesting that other patient-specific factors might influence the regenerative potential of MSCs to a greater extent than age2527). The present study cannot isolate age effects and the effects of acute ACL-injury and knee osteoarthritis, as all young donors were subject to knee injury and all old donors received a total knee replacement due to osteoarthritis.…”

“…Unlike other stem cell types, there is little research on the optimum culture conditions for LDSC survival and expansion. The survival and function of cells are normally dependent upon the culture conditions, and oxygen tension appears to have an effect on LDSC metabolism and matrix production [23]. In addition, certain media formulations and growth factors, such as basic fibroblast growth factor (bFGF) and transforming growth factor β 1 (TGF- β 1), promote stem cell proliferation and differentiation [24].…”

“…Hypoxia is the microenvironment of both tenocytes and MSCs. The oxygen tension has been shown to be 1-10% in tendon [16], 1-2% in bone marrow and approximately 3% in adipose tissue [17].…”
