Hypoxia Inducible Factor-2α Stabilization and Maxi-K + Channel β 1 -Subunit Gene Repression by Hypoxia in Cardiac Myocytes

Abstract: Abstract-The Ca 2ϩ -and voltage-dependent K ϩ (maxi-K) channel ␤ 1 -subunit mRNA is particularly abundant in cardiomyocytes but its functional role is unknown. This is intriguing because functional maxi-K channels are not found in cardiomyocyte plasmalemma, although they have been suggested to be in the inner mitochondrial membrane and participate in cardioprotection. We report here that ␤ 1 protein may interact with mitochondrial proteins and that the ␤ 1 -subunit gene (KCNMB1) is repressed by sustained hypox… Show more

“…Apparently, dsRNA injection is not applicable for pest control in the field, simpler and more convenient means of dsRNA delivery is necessary. It was reported that oral feeding with dsRNA causes RNAi effects in several insects [18], [19]. In a previous study, we showed that oral feeding with bacterially expressed dsRNA of a non-midgut gene, CHSA , in the beet armyworm, Spodoptera exigua , results in lethality [20].…”

“…Standard PCRs were performed using Biotaq TM DNA Polymerase (Bioline), in a T-professional Trio Thermocycler (Biometra). Primers used for the reactions amplified the following genes: P2X2 and P2X3 [64], P2Y12 [65], and cyclophilin as housekeeping [66]. Primer sequences and PCR product sizes are shown in the following table: Quantitative PCRs were performed using TaqMan specific probes for selected genes (Scn9a, Ntng1, Ascl1, Ncam2, Slc18a1, Il20ra, Pfkp, Pcsk6, and Hprt1) and the TaqMan Gene Expression Master Mix (Applied Biosystems).…”

“…For example, SIRT1 is able to induce the deacetylation and activation of Forkhead box O (FOXO1, 3 &4) [190,191], which regulates the expression of antioxidant enzymes and induces cardiac resistance to oxidative stress [192]. SIRT1 also deacetylates and stimulates Hif-2α [193], which is known to regulate cardiomyocyte resistance to ischemia [194] and PGC-1α [195], which promotes mitochondrial biogenesis and may be beneficial for recovery after MI [196]. Furthermore, SIRT1 inhibits the transcriptional activity of NF-kB via deacetylation of subunit RelA/p65 [197], which may prevent inflammatory responses during MI [198].…”
