Hypoxia and acidosis impair cGMP synthesis in microvascular coronary endothelial cells

Abstract: To characterize the effects of ischemia on cGMP synthesis in microvascular endothelium, cultured endothelial cells from adult rat hearts were exposed to hypoxia or normoxia at pH 6.4 or 7.4. Cellular cGMP and soluble (sGC) and membrane guanylyl cyclase (mGC) activities were measured after stimulation of sGC (S-nitroso-N-acetyl-penicillamine) or mGC (urodilatin) or after no stimulation. Cell death (lactate dehydrogenase release) was negligible in all experiments. Hypoxia at pH 6.4 induced a rapid approximately … Show more

“…[16][17][18] Simulated ischemia severely depresses cGMP synthesis in microvascular endothelium through ATP depletion and acidosis, thereby reducing NO production. 19 Hypoxic ischemia induces a combination of hyperosmolarity and lactic acidosis which modify red blood cells. The erythrocyte membrane becomes more rigid with time, which contributes to the decreased erythrocyte deformability after AMI.…”

“…Endothelial cells were obtained as previously described in detail. 28 To analyze cGMP in the whole hearts, hearts were pulverized under liquid nitrogen and homogenized in cold trichloroacetic acid at 7.5% (wt/vol). After centrifugation (14,000g, 15 minutes, 4°C), the supernatant was collected and washed with water-saturated diethylether.…”

“…cGMP content in myocytes and endothelial cells was assessed in ethanol extracts from the corresponding cell cultures, dried, and resuspended in acetate buffer (5 mmol/L, pH 4.8). cGMP was measured in the extracts by radioimmunoassay using acetylated [ 3 H]-cGMP as previously described, 28 and results were expressed as femtomoles of cGMP per milligram of protein.…”

“…Because acidosis often coexists with hypoxia, the role of acidosis as an independent modulator of vascular cell behavior has, until recently, been overlooked. However, recent studies in vitro and in vivo support the idea that EA has cellular effects independent of hypoxia (2,9,21,27,32,35).Heme oxygenase (HO) and its enzymatic products biliverdin and CO have received increasing recognition as biologically important modulators of cellular interactions in the vasculature. The inducible form of this enzyme, HO-1, is ubiquitously distributed in mammalian tissues, and its expression is regulated by a number of nonheme inducers such as cytokines, heavy metals, hormones and endotoxin (1).…”
