Hypotonically induced changes in the plasma membrane of cultured mammalian cells

Abstract: Cells from three cell lines were electrorotated in media of osmotic strengths from 330 mOsm to 60 mOsm. From the field-frequency dependence of the rotation speed, the passive electrical properties of the surfaces were deduced. In all cases, the area-specific membrane capacitance (Cm) decreased with osmolality. At 280 mOsm (iso-osmotic), SP2 (mouse myeloma) and G8 (hybridoma) cells had Cm values of 1.01 +/- 0.04 microF/cm2 and 1.09 +/- 0.03 microF/cm2, respectively, whereas dispase-treated L-cells (sarcoma fibr… Show more

“…The increase in cell size requires the unfolding of undulations and invaginations of cell membrane. 42,43 This decrease in membrane undulations results in the decrease of undulation repulsive forces, yielding better cell-cell contacts. 31,[44][45][46][47] Differences among cell lines were found in regulatory volume decrease dynamics.…”

“…The decrease of the mean C, value (and its CV) in hypotonically swollen cells is due to the disappearance of membrane folds and microvilli (12,19,37,38). At physiological osmolality (280-300 mOsm), large variations in the plasma membrane morphology cause the CV of the individual C , values to be higher.…”

“…The polarizability of living cells depends strongly on their composition, morphology, and phenotype and is also highly dependent on the frequency of the applied electrical field. 26,27 This means that for a given particle type and suspending medium, the particle can experience, at a certain ac frequency applied to the electrodes, a translational force directed toward regions of high electric field strength ͑this phenomenon is called positive DEP, i.e., pDEP͒. Alternatively, by simply changing the frequency, they may experience a force that will direct the particle away from the high electric field strength regions ͑this phenomenon is called negative DEP, i.e., nDEP͒.…”
