Hypothyroidism modulates beta adrenergic receptor adenylate cyclase interactions in rat reticulocytes.

Abstract: A B S T R A C T We have investigated alterations in beta adrenergic receptor binding sites of rat reticulocytes occurring in animals rendered hypothyroid by thyroidectomy. Beta adrenergic receptor interactions were assessed by measuring the number of (-)[3H]-dihydroalprenolol binding sites and the ability of an agonist to compete for occupancy of the receptors. The number of receptors was significantly reduced in cells from the hypothyroid animals. In addition, there were significant agonist-specific alteratio… Show more

“…Reticulocytes from hypothyroid rats, like the hypothyroid turkey erythrocytes described in the preceding section, have a reduction in /3 receptor number of approximately 50% (68). Although the K d for the radioligand [ 3 H]DHA is unchanged (68), it has been reported that there is a reduction in overall binding affinity for isoproterenol attributable to changes in the selective binding affinities of sites in both high-and lowaffinity states (68).…”

“…Rat adipose tissue BAR have been said to be unchanged in hyperthyroid states [15], and liver BAR decreased [22]. Conversely, the hypothyroid state is generally associated with reductions in BAR density in heart [6], turkey erythrocytes [3], rat reticulocytes [31], cortex [16] and cerebellum [29].…”

“…In other situations where N is genetically deficient, such as the cyc-variant of S49 lymphoma cells (22), or in situations where there is uncoupling of R from N such as desensitization (35), hypothyroidism (36), or hypoadrenalism (37), there is a reduction in the agonist-specific high affinity binding component that is demonstrated by a right-shifted and steepened agonist competition curve. This was precisely the situation documented in this study for the competition curves of the PHP-Ia patients.…”
