Hyperlipidemia represents a major risk factor for cardiovascular diseases leading to myocardial injury. The present study aimed to illustrate the myocardial injury induced in a diabetic hyperlipidemic rat model and the effect of vitamin D3, 10-DHGD intake either individually or in combination form. Male albino rats were selected for the study, received alloxan, hypercholesterolemic diet, and categorized into four groups. The first one (DHC), received hypercholesterolemic diet only and referred to as control. The remaining groups (2, 3, 4) received vitamin D3, 10-DHGD, and combination of both, respectively. Certain biomarkers that were selected for MI evaluation included blood glucose, lipogram pattern, Copeptin, C-reactive protein, myeloperoxidase, heart fatty acid-binding protein, and histopathological changes in myocardium and aorta. Vitamin D3 and 10-DHGD intake induced significant hypoglycemic, hypolipidemic effects, decreased inflammation, and MI biomarkers. Decreased myocardial vacuoles, foam cells, and intimal lesions were also observed compared to DHC. Their combination intake induced more marked reduction in all biomarkers and showed a histopathological pattern similar to normal features of myocardium and aorta. Our findings suggest the therapeutic roles of vitamin D3, 10-DHGD, and their combination against myocardial injury in diabetic hyperlipidemic rats. Impact statement Hyperlipidemia represents a major risk factor for cardiovascular diseases leading to myocardial injury (MI). Protective Effects of Borago officinalis (Borago) on Cold Restraint Stress-Induced Gastric Ulcers in Rats: A Pilot Study

Stress is a typical body's natural defense to a generic physical or psychic change. A specific linking mechanism between ulcer onset and psycho-physical stress prolonged exposure has been reported. We decided to investigate the possible effects of Borago officinalis L. (Borago) in preventing physical (stress)-induced gastric ulcers in a rat model. Eighty male Sprague-Dawley rats were randomly divided into 16 groups, pretreated with a control solution, omeprazole (20 mg/kg), Borago methanolic extract (25, 50, 100, 250, and 500 mg/kg), Borago organic extract (50, 100, 250, and 500 mg/kg), Borago aqueous extract (5, 10, 20, 30, and 40 mg/kg), and D(-)-2-Amino-5-phosphonovaleric acid (AP5) (25 mg/kg) and kept in stressful conditions such as water immersion and restraint-induced stress ulcers. The animals were sacrificed and their stomach scored for the severity and the number of gastric ulcers. Methanolic extract (500 mg/kg) significantly reduced both ulcer parameters ( *** p < 0.001 and ** p < 0.01, respectively). Aqueous and organic extract significantly decreased severity score at 5 and 10 mg/kg ( ** p < 0.01 and *** p < 0.001, respectively), and at 250 and 500 mg/kg ( *** p < 0.001), respectively, while gastric ulcers' resulted number significantly reduced only at 10 mg/kg ( * p < 0.05) and at 500 mg/kg ( ** p < 0.01), respectively. On the other hand, aqueous extract significantly increased the mucosal gastric content of cAMP ( * p < 0.05) and NR2A and NR2B subunits ( * p < 0.05 and ** p < 0.01, respectively) at 5 mg/kg. Organic extract showed also a significant cytotoxic effect at 500 and 1,000 mg/kg with a 3T3 cell viability reduction of 43.6% ( ** p < 0.01) and 92.1% ( *** p < 0.001), respectively. Borago aqueous extract at 10 mg/kg could be considered as a potential protective agent against stress-induced ulcers, and it is reasonable to possibly ascribe such protective activity to a modulation of the NR2A and NR2B subunit expression.

Introducción: El uso indiscriminado de agentes antiparasitarios ha resultado en el establecimiento de resistencia a ellos. Por lo cual es necesario el desarrollo de nuevas alternativas de tratamiento. Los productos naturales poseen diversas cualidades como posibles coadyuvantes en terapias contra distintos agentes etiológicos, entre los que destaca sus efectos antiparasitarios. Objetivo: Evaluar la actividad antiparasitaria, antioxidante, citotóxica y citoprotectora de Berberina (Ber), Curcumina (Cur) y Quercetina (Qr). Metodología: Se prepararon soluciones de Ber, Cur y Qr grado analítico y se realizaron alícuotas a diferentes concentraciones para su evaluación en contra de: Entamoeba histolytica, Trichomonas vaginalis y Strongyloides venezuelensis, para ello, se determinó la concentración inhibitoria media (IC50), además se determinó la capacidad antioxidante (CE50) mediante la prueba de DPPH, ambos por la prueba de Probit. Mediante la técnica de hemólisis se determinó la actividad citotóxica y citoprotectora, se aplicó Anova y la prueba de Tukey para determinar la diferencia de las medias en los tratamientos evaluados. Resultados: Ber, Cur y Qr, presentaron actividad en contra de E. histolytica, T. vaginalis y S. venezuelensis in-vitro. Ber presentó IC50 de 1.7, 1.2 y 1.9 μM respectivamente siendo más efectivo en comparación de Cur con IC50 de 55.3, 40.6 y 13.7 μM o Qr con IC50 de 147.2, 93.2 y 110.9 μM, sin embargo, la mejor actividad antioxidante (EC50 = 1.1 μg/ml), citoprotectora y menos hemolítica, fue presentada por Qr (P < 0.001) en comparación con el control evaluado. Conclusiones: Los metabolitos de origen natural berberina, curcumina y quercetina, poseen actividad en contra de trofozoítos de E. histolytica, T. vaginalis y larvas de S. venezuelensis en dosis bajas comparables con los fármacos de referencia para el caso de Ber. Además, estos productos de origen natural, no sintético podrían ser objeto de futuras investigaciones para coadyuvar al tratamiento de parasitosis, ya que, en dosis bajas, mostraron actividad antioxidante sin mostrar hemólisis considerable en eritrocitos humanos.
