Hyperoxia Increases Protein Mass of 5-Lipoxygenase and Its Activating Protein, Flap, and Leukotriene B 4 Output in Newborn Rat Lungs

Abstract: In this study, the authors examined in newborn rat lung tissues the release of leukotriene B(4) (LTB(4)) from tissue explants in vitro, the protein expression of the LT-synthesizing enzyme, 5-lipoxygenase (5-LO), and its activating protein (FLAP), and the effects of in vivo hyperoxic exposure (>95% O(2)) on these parameters. Basal LTB(4) output increased from 0.98 ng/mgDNA/30 min at day 1 to 3.3 ng/mgDNA/30 min at day 28 (P <.05). Exposure of rat pups to >95% O(2) from days 1 to 7 and 60% O(2) from days 7 to 2… Show more

“…Most LMs are produced by the combined reactions of regio‐ and stereo‐specific LOXs determined by the oxygen migration in the protein matrix (Coffa & Brash, 2004; Mishra & Mishra, 2019, 2020; Saam et al, 2007). LOXs were discovered as 5 S ‐ (Gilbert et al, 2011; Hosford et al, 2002), 8 S / R ‐ (Gilbert et al, 2018; Okazaki et al, 2002), 11 S/R ‐ (An et al, 2018; Mortimer et al, 2006), 12 S / R ‐ (Deng et al, 2014; Horn et al, 2013; Jobard et al, 2002), and 15 S/R ‐types (Hamberg et al, 1998; Zhao et al, 2011) on ARA in animals and plants, but rarely in microorganisms.…”

“…Little is known about the effect of ROI on the expression of 5-LO. Other investigators examined the effect of hyperoxia on 5-LO expression in the lungs of newborn rat lung tissue [43]. LT synthesis and total lung expression of 5-LO were increased in animals treated with hyperoxia.…”

“…As products of cyclooxygenase and lipoxygenase, these oxylipins may result in abnormal inflammatory responses in infants who develop BPD. Indeed, hyperoxia increased the activity and levels of cyclooxygenase and lipoxygenase in newborn rat lungs, and deficiency of cyclooxygenase 2 attenuated hyperoxia-induced lung inflammatory responses [ 82 , 83 ]. Therefore, reducing lipid peroxidation and oxylipins could be a potential therapy to prevent or mitigate the development of BPD.…”
