Hydroxyurea in Thalassemia Major

Abstract: BACKGROUNDHydroxyurea (HU) is found to be effective in sickle cell anemia and thalassemia intermedia. Its usefulness in thalassemia major is not clearly defined. The aim of the study was to analyze effectiveness of hydroxyurea in children with beta thalassemia major.

“…Hydroxyurea showed no serious side effects in almost all the studies conducted so far, except mild liver or renal impairment observed in a couple of researches [12][13][14][15][16][17][18]. It can be used safely in children as a single oral dose per day and monitored with complete blood count, alanine aminotransferase (ALT), and serum creatinine levels regularly.…”

“…Yadav A et al studied the efficacy of hydroxyurea in 25 cases of transfusion-dependent thalassemia-major. Hydroxyurea significantly increased mean hemoglobin levels and reduced the number of transfusions per day with no significant reportable side effects of therapy [ 15 ]. Ghosh d et al studied the effect of hydroxyurea on 49 transfusion-dependent HbE/beta-thalassemia major cases and found significant improvement in the transfusion-free interval [ 16 ].…”

“…Sources: References [14,17] Three studies compared the change in HbF between two groups. Pooled meta-analysis showed that the mean HbF was higher in patients receiving hydroxyurea; however, the difference was statistically insignificant (MD: 2.24, 95% CI: -0.53, 5.00), as shown in Figure 4.…”
