Hydroxyl regioisomerization of anthracycline catalyzed by a four-enzyme cascade

Abstract: Ranking among the most effective anticancer drugs, anthracyclines represent an important family of aromatic polyketides generated by type II polyketide synthases (PKSs). After formation of polyketide cores, the post-PKS tailoring modifications endow the scaffold with various structural diversities and biological activities. Here we demonstrate an unprecedented four-enzyme-participated hydroxyl regioisomerization process involved in the biosynthesis of kosinostatin. First, KstA15 and KstA16 function together to… Show more

“…Total extraction of the pigment from mycelia was obtained only with ethanol. After ethanol evaporation, the extracted pigment is not water soluble in agreement with the case of prodigiosin [30] or aglycone moiety of other anthracyclines [31,32]. Curiously, part of it could acquire water soluble ability after air dry in fume hood as indicated for the case of strain MFB21 pigment ( Figure 5) which showed 36% of soluble pigment after 24 h air dry.…”

“…Various reports previously summarized that anthracycline antibiotics biosynthesized by actinomycetes have numerously shown cytotoxicity against human tumors or tumor cell lines (Metsä-Ketelä et al, 2007;Zhang et al, 2017). Therefore, anthracycline antibiotics from actinomycetes have been so far, one of the most effective substances widely used in the treatment of many types of cancer (Minotti et al, 2004).…”

“…Gene duplication and catalytic promiscuity have been important in the evolution of the unique substructure. This chemistry originates from an unusual two‐component mono‐oxygenase system that is responsible for the installation of the C1 hydroxyl group . The system is composed of an SDR‐enzyme, SnoaW, and a protein SnoaL2 homologous to canonical polyketide cyclases such as SnoaL .…”
