Hydroxycarboxylic Acid Receptor Ligands Modulate Proinflammatory Cytokine Expression in Human Macrophages and Adipocytes without Affecting Adipose Differentiation

Abstract: Members of the hydroxycarboxylic acid receptor (HCA1-3) family are mainly expressed in adipocytes and immune cells. HCA2 ligand, niacin, has been used for decades as lipid-modifying drug. Recent studies suggest that HCA ligands can be involved in the modulation of inflammatory processes. In this study, we evaluated the effects of HCA1-3 ligands on adipose differentiation and cytokine expression in human adipocytes and macrophages. Simpson-Golabi-Behmel syndrome (SGBS) preadipocytes were induced to differentiat… Show more

“…Furthermore, our findings that GPR81 is significantly reduced upon exposure to OSS suggest the importance of this receptor in endothelial cell protection. A recent study involving adipocytes and THP‐1 macrophages demonstrated that lactate‐mediated activation of GPR81 significantly reduced expression of IL‐6 and IL‐8 induced by lipopolysaccharide (LPS) . Another recent study suggests that HCA receptors, including GPR81 (HCA 1 ) may serve as therapeutic targets against inflammatory diseases .…”

“…Moreover, chemotaxis of neutrophils and monocytes upon HCA 3 activation has been reported [25,26]. However, HCA 3 is rather suggested to elicit hypo-responsiveness of the immune system through mediation of anti-inflammatory processes [25,28].…”

“…Similar to the mediumchain fatty acid receptor, HCA3 is expressed on immune cells. However, unlike GPR84, activation of HCA3 may also suppress the production of proinflammatory cytokines in response to LPS [4]. Thus, we can assume that capric acid, acting through cognate receptor, is capable of shifting the Th1/Th2 ratio towards the prevalence of Th2-lymphocytes.…”
