Hydroxy- and Amino-Phosphonates and -Bisphosphonates: Synthetic Methods and Their Biological Applications

Abstract: Phosphonates and bisphosphonates are stable analogs of phosphates and pyrophosphates that are characterized by one and two carbon–phosphorus bonds, respectively. Among the various phosphonates and bisphosphonates, hydroxy and amino substitutes are of interest as effective in medicinal and industrial chemistry. For example, hydroxy bisphosphonates have proven to be effective for the prevention of bone loss, especially in osteoporotic disease. On the other hand, different substitutions on the carbon atom connect… Show more

“…The first BTAs approved by the FDA for the treatment of bone metastases were bisphosphonates. Apart from the treatment of bone metastases, bisphosphonates are commonly applied in the therapy of osteoporosis and Paget’s disease [ 147 ]. Currently, they are also investigated as a potential treatment for otosclerosis, osteoarthritis of the knee, inflammatory rheumatic diseases, and many others diseases [ 148 , 149 , 150 ]…”

“…They have a wide range of applications, such as in (pro)drugs in medicine, among others. The presence of strong P–C bonds also makes them resistant to hydrolysis, allowing their use in the treatment of disorders of the calcium metabolism and bone diseases [ 1 , 2 , 3 , 4 , 5 , 6 ].…”

“…L carnitine ( Figure 1 ). This modification confers greater solubility, stability, and potency ( Kaboudin et al, 2022 ). β-HPC was proposed for the treatment of metabolic syndrome because, in animal models, it reduced blood levels of insulin, triglycerides and cholesterol in the liver and serum ( Gómez-Solís et al, 2011 ; Reyes-Esparza et al, 2014 ; De la Cruz-Cordero et al, 2016 ).…”
