In this review, method development for hydrophilic interaction LC (HILIC) is highlighted. HILIC is a chromatographic technique that uses aqueous-organic solvent mobile phases with a high organic-solvent fraction, and a hydrophilic stationary phase. It is mainly applied for the separation of polar and hydrophilic compounds. Method development, in general, can be done uni- or multivariately. In the univariate approach, the factors that are expected to potentially affect the separation of the compounds will be examined sequentially and one-at-a-time. All HILIC methods found in the literature were developed in this way. For these methods, the analytes, the considered factors, the selected responses, and the finally chosen experimental conditions are discussed in this review. Where examined, the method validation and the comparison with other analytical assay methods is also described. For the multivariate method-development approach, which is based on the use of experimental designs, only a possible strategy is presented, because of the lack of relevant publications in the literature.

A highly selective micellar electrokinetic capillary chromatography (MEKC) method with laser-induced fluorescence (LIF) detection for the sensitive determination of pheomelanin in diverse biological materials was originally described. The derivatization reagent, 4-(1-pyrene) butyric acid N-hydroxysuccinimide ester (PSE), allowed for the selective detection of the two aminohydroxyphenylalanines (AHPs) markers for pheomelanin monitored at 500 nm. Multiple labeling of two AHPs with PSE allowed the formation of intramolecular excimers that emit at longer wavelengths (500 nm) than the mono-labeled analytes (360-420 nm) based on intramolecular excimerforming fluorescence derivatization. Optimal separation of the labeled polyamines was achieved using a separation buffer consisting of 20 mM phosphate pH 7.4, 30 mM cholate, and 30% methanol. Using these conditions, the two AHPs were separated within 12 min, and the relative standard deviations (RSDs) were less than 1.5 and 1.6% (intra-run), 3.8 and 4.6% (inter-run, for a 6-day period) for the migration times and peak areas (n = 10), respectively. This method was successfully applied to the monitoring of pheomelanin in diverse biological samples with the spiked recoveries in the range of 94-101%. At a signal-to-noise ratio of 3, the detection limit for AHPs in the real samples was 31 pM for 3-AHP and 35 pM for 4-AHP, respectively, which are superior to those previously reported in the literature using fluorescence detection.

A new HPLC method was developed and validated for the determination of asymmetric and symmetric dimethylarginines and l-arginine in human plasma. After SPE and evaporation of the eluate, the samples were derivatised with an o-phthaldialdehyde reagent containing 3-mercaptopropionic acid. The derivatives formed were analysed by isocratic RP-HPLC with electrochemical detection at +320 mV. The mobile phase consisted of 50 mM phosphate buffer (pH 6.1) containing 10% v/v acetonitrile, the flow rate was 1 mL/min. The retention times of all compounds including monomethylarginine (internal standard) were <24 min. The LODs (S/N 3:1) were 0.012 μM for both dimethylarginines and 0.013 μM for L-arginine; the linearity of the method was from 0.1 to 20 μM for both dimethylarginines and from 1 to 200 μM for L-arginine. Absolute extraction recoveries measured for all analytes ranged from 85 to 88%.
