Hydrogen sulfide producing enzymes in pregnancy and preeclampsia

Abstract: Preeclampsia, a human pregnancy specific disorder is characterized by an anti-angiogenic state. As hydrogen sulfide (H(2)S) has pro-angiogenic and anti-oxidative characteristics, we hypothesized that H(2)S levels could play a role in the pathogenesis of preeclampsia and studied the placental expression of the H(2)S-producing enzymes cystathionine-γ-lyase (CSE) and cystathionine-β-synthase (CBS). CBS and CSE protein are expressed in the fetal-placental endothelium and CBS only in Hofbauer cells. CBS mRNA expres… Show more

“…52 Interestingly, the expression of CBS and CSE in women with preeclampsia is decreased in the placenta. 47,48 The administration of H 2 S had the same effect on sFlt1 levels in pregnant rats, compared with nonpregnant rats (data not shown). Importantly, pups from NaHS-treated mothers showed no apparent dysmorphic characteristics and there were no adverse effects of NaHS on placental and pup weight (Supplemental Figure 3).…”

“…Clinically, the rats showed decreased proteinuria, hypertension, and glomerular injury (38). Conversely, decreased levels of the precursor molecules of H 2 S have been found in patients with preeclampsia (37,39,40). Chronic administration of a cystathionine g-lyase inhibitor, DL-propargylglycine, to pregnant mice resulted in elevated mean BP, liver damage, and decreased fetal growth.…”

“…10,33 In the placenta, these enzymes are active (both rat and human) and are able to produce H 2 S. 6 The catalytic activity of placental CBS was also shown by incubation of human placental explants with increasing homocysteine concentrations leading to elevated CBS activity and increased cysteine in vitro. 34 In the human fetoplacental circulation, H 2 S mediates vasodilatation in vitro via K ATP channels and by interaction with NO.…”
